摘要
目的:建立野生和栽培秦艽药材的HPLC指纹图谱,结合化学模式识别方法对两者进行判别,为不同来源秦艽药材鉴别和质量评价提供依据。方法:利用HPLC法,测定18批不同产地野生与栽培秦艽药材的HPLC指纹图谱,建立指纹图谱共有模式,采用中药色谱指纹图谱相似度评价系统(2012A版)计算相似度,运用聚类分析(HCA)和最小偏二乘法-判别分析(PLS-DA)对其进行模式识别研究。结果:建立了野生与栽培秦艽药材HPLC指纹图谱共有模式,标定了19个共有峰,18批不同产地野生与栽培秦艽药材指纹图谱与对照指纹图谱的相似度均>0.93。HCA、PLS-DA能将野生与栽培秦艽药材明确区分,马钱苷酸、獐牙菜苷、6’-O-β-D-葡萄糖基龙胆苦苷、獐牙菜苦苷等7个成分是导致二者差异的主要标记物。结论:建立的秦艽药材指纹图谱特征性强,结合化学模式识别方法可有效评价秦艽药材质量以及区分其野生与栽培品,为秦艽药材的质量控制和化学计量分类提供参考依据。
Objective: To establish the HPLC fingerprint of wild and cultivated Gentiana macrophylla Pall,and recognize them according to the chemical pattern in the expectation of providing the basis for the quality control and domestication cultivation of Gentiana macrophylla Pall of origins. Methods: Eighteen samples of wild and cultivated Gentiana macrophylla Pall collected from different origins were detected by HPLC,and a common mode of fingerprint was established.The Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012 A edition) was used to evaluate the similarity of samples.The differences among the samples were identified by chemical pattern recognition methods including hierarchical cluster analysis(HCA) and partial leastsquares discriminate analysis(PLS-DA).Results: The HPLC fingerprint of Gentiana macrophylla Pall was obtained,and 19 common peaks were found in the chromatogram.The similarities of all samples were over 0.93,and HCA and PLS-DA results demonstrated obvious distinctions between wild and cultivated Gentiana macrophylla Pall.Seven constituents,such as loganic acid,sweroside,6’-O-β-D-glucosyl gentiopicroside and swertiamarin,were identified as biomarkers representing major differences between the two varieties.Conclusion: The HPLC chromatogram of Gentiana macrophylla Pall developed in this paper has obvious characteristics. After combined with the pattern recognition mode,it can be used as an effective method for evaluating the quality of Gentiana macrophylla Pall and distinguishing wild and cultivated Gentiana macrophylla Pall,which provides reference for the quality control and domestication introduction of Gentiana macrophylla Pall.
