基于网络药理学探讨大柴胡汤对2型糖尿病的治疗作用机制 被引量:8

摘要 目的运用网络药理学的方法探讨大柴胡汤治疗2型糖尿病的作用机制,对作用靶点和信号通路进行预测和筛选。方法首先在SymMap中医药生物信息学平台获取大柴胡汤的潜在作用靶点信息,在CTD疾病数据库获取2型糖尿病的潜在治疗靶点,将两者取交集即得筛选出的药物疾病靶点;使用STRING构建靶点的PPI网络;运用CluGO工具对靶点进行GO功能和KEGG信号通路富集分析,并筛选大柴胡汤治疗糖尿病的活性化学成分。结果获取大柴胡汤靶点476个,其中与2型糖尿病相关联的靶点有41个,主要参与了葡萄糖跨膜转运、调节胰岛素分泌、葡萄糖稳态、胰岛素受体信号通路的调节等生物过程;还参与调控了胰岛素抵抗、青少年的成年人型糖尿病、2型糖尿病、胰岛素信号通路等相关通路。结论通过网络药理学分析显示,大柴胡汤对2型糖尿病的治疗作用可能与直接参与调节胰岛素的分泌相关作用途径有关。 Objective To explore the mechanisms of Dachaihu decoction in the treatment of type 2 diabetes by network pharmacology,and to predict and screen the targets and signal pathways.Methods The potential targets information of Dachaihu decoction was obtained from SymMap TCM bioinformatics platform,and the potential therapeutic targets of type 2 diabetes were obtained from CTD disease database.A targets’ PPI network was built by using STRING.GO and KEGG enrichment analyses on the targets were carried out by using CluGO tool.Besides,Dachaihu decoction was screened for the active chemicals in the treatment of diabetes.Results In total,476 targets of Dachaihu decoction were obtained,of which 41 targets were associated with type 2 diabetes.They were mainly involved in glucose transmembrane transport,regulating insulin secretion,glucose homeostasis,and regulating insulin receptor signaling pathways.They also participate in regulating insulin resistance,adolescent adult type diabetes,type 2 diabetes,insulin signaling pathway and other related pathways.Conclusion The analysis of network pharmacology shows that the therapeutic effects of Dachaihu decoction in the treatment of type 2 diabetes may be directly related to the pathways involved in regulating insulin secretion.
