补肾止颤方对6-OHDA诱导的帕金森病模型大鼠神经保护作用的研究 被引量:7

Study on the Protective Effects of Bushen Zhichan Prescription on Dopaminergic Neurons in 6-hydroxydopamine-induced Parkinsons Disease Model

摘要 目的探讨补肾止颤方对帕金森病(Parkinsons disease,PD)模型大鼠多巴胺能神经元的保护作用。方法将100只Sprague-Dawley大鼠随机分为5组:正常组、模型组、补肾止颤方低剂量组、中剂量组、高剂量组,补肾止颤方按照不同剂量溶于生理盐水中,每天中午灌胃1次,正常组和模型组予以相同体积的生理盐水灌胃。分别在处理10天和20天后检测各组大鼠的行为学,免疫组织化学法检测脑内多巴胺能神经元标志物酶酪氨酸羟化酶(Tyrosine hydroxylase,TH)和多巴胺递质转运子(Dopamine transmitter transporter,DAT),酶联免疫吸附测定(Enzyme-linked immunosorbent assay,ELISA)检测脑内多巴胺及其代谢产物的变化。结果和模型组比不同剂量的补肾止颤方均能够改善PD大鼠的行为学,提高TH、DAT、3,4二羟基苯乙酸(3,4 dihydroxyphenylacetic acid,DOPAC)、5羟色胺(Serotonin,5-HT)、高香草酸(High vanilla acid,HVA)的表达(P <0.05或P <0.01),同时下调单胺氧化酶B(Monoamine oxidase B,MAO-B)的表达(P <0.01)。结论补肾止颤方能够改善6-羟基多巴胺(6-hydroxydopamine,6-OHDA)损伤早期大鼠的行为学改变,增加帕金森病大鼠多巴胺能神经元的数量,并能维持多巴胺能神经元功能,降低神经损伤,有较好的神经保护作用。 Objective To explore the protective effects of Bushen Zhichan Prescription(BZP) on dopaminergic neurons in Parkinson’s disease(PD) model.Methods One hundred male Sprague-Dawley rats were randomly divided into 5 groups:normal group,model group,low-dose group,middle-dose group,and high-dose group.BZP was dissolved at different doses in normal saline,and gavage was given once a day at noon.The normal group and the model group were given the same volume of normal saline.After 10 and 20 days of treatment,the behavior of PD rats in each group was observed,the brain dopaminergic neuronal markers dopamine transmitter transporter(DAT) and tyrosine hydroxylase(TH) were detected by immunohistochemistry,and the changes of the metabolites of Dopaminergic Neurons were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with the model group,different doses of BZP can improve the behavior of PD rats,and improve TH,DAT,3,4 dihydroxyphenylacetic acid(DOPAC),serotonin(5-HT),and high vanilla acid(HVA)(P <0.05 or P <0.01),while the expression of monoamine oxidase B(MAO-B) was simultaneously down-regulated(P <0.01).Conclusion BZP can improve the behavioral changes of rats with early 6-OHDA injury,increase the number of dopaminergic neurons in PD rats,maintain the function of dopaminergic neurons,and reduce nerve injury.All of these demonstrated that BZP has neuroprotective effects.
