隐丹参酮对小鼠变应性接触性皮炎的影响及作用机制 被引量:2

摘要 [目的]探讨隐丹参酮对小鼠变应性接触性皮炎(ACD)的影响,并观察其机制是否与Th1,Th2型细胞炎性因子及MAPKs蛋白的作用有关.[方法]取BALB/c小鼠随机分为对照组、模型组、隐丹参酮低、高剂量(20,40 mg/kg)组及地塞米松(1 mg/kg)组,其中模型组、隐丹参酮低、高剂量组采用2,4-二硝基氯苯诱导小鼠出现ACD,隐丹参酮低、高剂量组在致敏成功后给药.观察各组小鼠激发后皮肤形态学改变、组织病理学改变、小鼠脾指数、血清中白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)的变化及皮肤组织中MAPKs蛋白的表达情况.[结果]与模型组比较,隐丹参酮低、高剂量组小鼠皮肤损伤严重程度明显减轻,炎症组织细胞浸润明显减少,脾指数明显降低,血清中IL-4,IFN-γ及皮肤中MAPKs蛋白表达水平均显著降低,相比较差异均有统计学意义(P<0.05,P<0.01).[结论]隐丹参酮对小鼠ACD具有治疗作用,其机制认为可能与隐丹参酮调节Th1,Th2型细胞炎性因子及抑制MAPKs蛋白发挥作用有关系. OBJECTIVE To explore the effects of cryptotanshinone on allergic contact dermatitis(ACD)in mice,and to observe whether the mechanism was related to the effects of inflammatory factors in Th1 and Th2 cells and MAPKs protein.METHODS BALB/c mice were randomly divided into 5 groups,including control group,model group,low and high-dose of cryptotanshinone group(20,40 mg/kg),and dexamethasone group(1 mg/kg).ACD was induced by 2,4-dinitrochlorobenzene in mice of the model group,the low and high-dose group.The low and high-dose of cryptotanshinone groups were given the cryptotanshinone after successful sensitization.The morphological changes of the skin,histopathological changes,mice spleen index,serum interleukin-4(IL-4),interferon-γ(IFN-γ)changes and MAPKs protein expression of skin tissues were observed after stimulation in each group.RESULTS Compared with the model group,the severity of skin injury,inflammatory cell infiltration,spleen index,serum IL-4,IFN-γand skin MAPKs protein levels in the low and high-dose of cryptotanshinone group were significantly reduced,and the differences were statistically significant(P<0.05,P<0.01).CONCLUSION Cryptotanshinone has a therapeutic effect on ACD in mice,and its mechanism might be related to the regulation of inflammatory factors in Th1 and Th2 cells and the inhibition of MAPKs protein.
