3D打印PLA-nHA复合支架膜在犬牙周组织再生中的应用 被引量:4

摘要 目的探讨3D打印不同比例聚乳酸(polylactic acid,PLA)-纳米羟基磷灰石(nanohydroxyapatite,nHA)复合支架膜进行引导组织再生术(guided tissue regeneration,GTR)后犬牙周组织修复的情况。方法3D打印不同比例的PLA-nPLA复合支架膜,分别将不同组屏障膜[对照组、实验1组(15%nHA:85%PLA)、实验2组(30%nHA:70%PLA)、实验3组(50%nHA:50%PLA)、实验4组(75%nHA:25%PLA)、实验5组(100%PLA)、实验6组(100%nHA)及空白组]进行细胞毒性检测,并将细胞毒性合格的复合支架膜植入牙周病犬术区行GTR术,空白组仅于术后缝合术区,不置屏障膜。于术后12周末处死动物,制取下颌骨标本后进行组织学检测。结果3D打印不同比例的复合支架膜均无明显细胞毒性,免疫组化结果显示:对照组骨涎蛋白及I型胶原蛋白显示强阳性,实验6组及实验4组次之,实验3组、实验2组、实验1组、实验5组、空白组依次降低,实验5组与空白组相比,阳性强度差异不大。结论胶原膜和比例为(75%nHA:25%PLA)复合支架膜在进行犬牙周GTR术后效果较好,有进一步的研究价值。 Objective To compare 3D printing of different ratios of PLA-nPLA composite scaffold membrane in periodontal disease dogs after guided tissue regeneration(GTR)canine periodontal tissue repair.Methods 3D printed different ratios of PLA-nPLA composite scaffold membranes,the different groups of barrier membranes[control group,experiment group 1(15%nHA:85%PLA),experiment group 2(30%nHA∶70%PLA),experiment group 3(50%nHA∶50%PLA),experiment group 4(75%nHA∶25%PLA),experiment group 5(100%PLA),experiment group 6(100%nHA)and blank group]were tested for cytotoxicity,and implanted in the periodontal disease dog area for GTR.The blank group was only in the postoperative suture area without barrier film.The animals were sacrificed 12 weeks after the operation,and the mandibles were prepared for histological examination.Results 3D printed composite scaffold membranes with no significant cytotoxicity.Immunohistochemical results suggest:bone sialoprotein and type Ⅰ collagen of collagen membrane group showed strong positive results,the experiment group 6 and experiment group 4 followed,the experimental group 3,the experimental group 2,the experimental group 1,the experimental group 5,and the blank control group are decreased in order.Compared with the blank group,the positive intensity of the experiment group 5 was not significantly different.Conclusion The collagen membrane and the ratio of(75%nHA∶25%PLA)composite scaffold membrane have better effect after canine periodontal GTR,which has further research value.
