痹玉康Ⅰ号干预CIA模型大鼠关节炎症病理损伤的实验研究

Experimental Study of Biyukang MixtureⅠIntervention on the Pathological Injury of Joint Inflammation in CIA Model Rats

摘要 目的探索痹玉康Ⅰ号对Ⅱ型胶原诱导的大鼠关节炎模型(CIA模型)大鼠关节炎症的影响及对滑膜炎症病理损伤的干预作用。方法建立CIA模型,造模成功的大鼠随机分为模型组、阳性药组、痹玉康Ⅰ号组,另设正常组,每组6只,连续干预4周。每周测量大鼠踝关节纵横径、足垫厚度,评估四肢关节肿胀程度。干预治疗结束后,比较各组踝关节肿胀程度,镜下观察大鼠双侧踝关节软骨组织及滑膜组织的形态学变化。结果与正常组比较,模型组大鼠双侧踝关节的横竖径以及足垫厚度测量值显著增加(P<0.05);与模型组比较,痹玉康Ⅰ号组横竖径、足垫厚度在7~28 d不同时间段均降低,在研究终点时显著下降(P<0.05)。病理形态学结果显示,痹玉康Ⅰ号组大鼠双侧踝关节病理损伤较模型组减轻,滑膜及软骨病理半定量评分降低(P<0.05),差异具有统计学意义。结论痹玉康Ⅰ号能够减轻CIA模型大鼠关节肿胀及病理损伤。 Objective To explore the effect of Biyukang MixtureⅠ(BMⅠ)on the arthritis of rats induced by typeⅡcollagen(CIA model)and its intervention on the pathological damage of synovial inflammation.Methods The CIA model was established.The rats that were successfully modeled were randomly divided into model group,positive drug group,BMI group,and a normal control group with 6 rats in each group.All interventions lasted for 4 successive weeks.The vertical and horizontal diameters of the ankle joints and the thickness of the foot pads were measured weekly to evaluate the swelling of the limbs and joints.After the intervention treatment,the degree of swelling of the ankle joints in each group was compared,and the histomorphological changes of the cartilage tissue and synovial tissue of the bilateral ankle joints of the rats were observed under the microscope.Result Compared with the normal group,the measured values of the transverse and vertical diameters of the bilateral ankle joints and the thickness of the foot pads in the model group increased significantly(P<0.05);compared with the model group,the transverse and vertical diameters and the thickness of the foot pads in the Biyukang group were It decreased in different time periods from 7 to 28 days,and it decreased significantly at the end of the study(P<0.05).The pathomorphology results showed that the bilateral ankle joint pathological damage of the Biyukang group was less than that of the model group,and the semi-quantitative score of the synovial and cartilage pathology was lower(P<0.05),and the difference was statistically significant.Conclusion BMI can reduce joint swelling and pathological injury in CIA model rats.
