壮督驱寒合剂对强直性脊柱炎小鼠Wnt3a、β-catenin表达及炎症因子的影响 被引量:2

Effect of Zhuangdu Quhan Mixture on Wnt3a,β-catenin Expression and Inflammatory Factors in Mice with Ankylosing Spondylitis

摘要 目的观察壮督驱寒合剂对强直性脊柱炎(AS)小鼠Wnt3a、β-连环蛋白(β-catenin)表达及炎症因子的影响。方法将60只雌性BALB/c小鼠随机分成空白组、模型组、塞来昔布组、中药低剂量组和中药高剂量组。除空白组外,其余各组用蛋白聚糖法诱导AS小鼠模型。造模成功后,塞来昔布组予塞来昔布灌胃,中药低剂量组和中药高剂量组分别予低剂量和高剂量中药灌胃,其余组灌胃等量生理盐水。每天1次,连续用药8周。比较各组之间滑膜Wnt3a和β-catenin mRNA和蛋白表达、血清肿瘤坏死因子α(TNF-α)和转化生长因子β1(TGF-β1)水平情况。结果与空白组比较,模型组滑膜组织Wnt3a、β-catenin mRNA和蛋白表达显著升高(P<0.01);与模型组比较,各治疗组Wnt3a、β-catenin mRNA和蛋白表达显著降低(P<0.01),其中中药高剂量组优于中药低剂量组(P<0.01),中药低剂量组优于塞来昔布组(P<0.05)。与空白组比较,模型组小鼠血清TNF-α、TGF-β1显著升高(P<0.01);与模型组比较,各治疗组TNF-α、TGF-β1显著降低(P<0.01),其中塞来昔布组优于中药高剂量组(P<0.05),中药高剂量组优于中药低剂量组(P<0.05)。结论壮督驱寒合剂对AS小鼠病理性成骨及炎症皆有一定的改善作用,其机制可能与调节Wnt/β-catenin信号通路Wnt3a、β-catenin蛋白表达,抑制TNF-α、TGF-β1水平有关。 Objective To observe the effects of Zhuangdu Quhan Mixture on Wnt3 a,β-catenin expression and inflammatory factors in mice with ankylosing spondylitis(AS).Methods 60 Sixty female BALB/c mice were randomly divided into blank group,model group,celecoxib group,low-dose Chinese medicine group and high-dose Chinese medicine group.Except for the blank group,the other groups used proteoglycan method to induce AS mouse models.After successful modeling,the celecoxib group was given intragastric administration of celecoxib,the low-dose Chinese medicine group and the high-dose Chinese medicine group were given low-dose and high-dose Chinese medicine respectively,and the other groups were given the same amount of normal saline.Take the medicine once a day for 8 weeks.The mRNA and protein expression of synovial Wnt3 a andβ-catenin,serum inflammatory factor tumor necrosis factorα(TNF-α)and transforming growth factorβ1(TGF-β1)levels were compared between the groups.Result Compared with the blank group,the mRNA and protein expressions of Wnt3 a andβ-catenin in the synovial tissues of the model group were significantly increased(P<0.01);compared with the model group,the mRNA and protein expressions of Wnt3 a andβ-catenin were significantly decreased in each treatment group(P<0.01),the high-dose group of Chinese medicine is better than the low-dose group(P﹤0.01),and the low-dose group of Chinese medicine is better than the celecoxib group(P<0.05).Compared with the blank group,the serum inflammatory factors TNF-αand TGF-β1 in the model group were significantly increased(P<0.01);compared with the model group,TNF-αand TGF-β1 in each treatment group were significantly decreased(P<0.01),the celecoxib group is better than the high-dose Chinese medicine group(P<0.05),and the high-dose Chinese medicine group is better than the low-dose group(P<0.05).Conclusion Zhuangduquhan mixture can improve the pathological osteogenesis and inflammation of AS mice,and its mechanism may be related to the regulation of Wnt/β-catenin signaling pathway Wnt3 a andβ-catenin protein expression,as well as the inhibition of TNF-αand TGF-β1 inflammatory factor levels.
