基于骨髓间充质干细胞的增殖表达探讨变应性鼻炎的体质基础

Study on the constitution basis of allergic rhinitis based on the proliferation of bone marrow mesenchymal stem cells

目的研究不同体质变应性鼻炎大鼠骨髓间充质干细胞增殖表达的规律。方法构建寒、热、常体大鼠变应性鼻炎模型,各体质分为干预和非干预组,寒体干预组和常体干预组喂饲附子理中汤,热体干预组喂饲黄连解毒汤,无干预组不做任何干预。在模型建立的第0,1,3,7天分别取材检测骨髓间充质干细胞的含量。结果第0天,寒体2组临床症状评分均明显高于热体2组、常体2组(P均<0.05),常体2组与热体2组间比较差异均无统计学意义(P均>0.05)。寒、热、常体无干预组实验第1,3,7天临床症状评分与实验当天比较差异均无统计学意义(P均>0.05);寒、热、常体干预组实验第7天临床症状评分均明显低于实验当天(P均<0.05)。寒体干预组、常体干预组实验第1天、第3天、第7天骨髓间充质干细胞表达量均明显高于同一时间点同体质非干预组(P均<0.05);热体干预组和非干预组实验第1天骨髓间充质干细胞表达量比较差异无统计学意义(P>0.05),实验第3天、第7天热体干预组均明显高于热体非干预组(P均<0.05)。经混合效应模型分析,非干预各组骨髓间充质干细胞表达量明显低于干预各组(P均<0.05);不同体质间比较,热体组和常体组间骨髓间充质干细胞表达量比较差异无统计学意义(P>0.05);而寒体组骨髓间充质干细胞表达量明显少于常体组和热体组(P均<0.05)。寒、热、常体无干预组的骨髓间充质干细胞增殖趋势整体上呈逐渐减少趋势,而干预组的骨髓间充质干细胞增殖趋势整体上呈先减少后增多趋势。结论骨髓间充质干细胞的增殖能反映变应性鼻炎的体质情况,寒体最容易发生变应性鼻炎,其骨髓间充质干细胞增殖最少,而体质治疗可以促进骨髓间充质干细胞的增殖。

Objective It is to study the proliferation of bone marrow mesenchymal stem cells(BMSCs)in rats with allergic rhinitis of different constitution.Methods The disease models of allergic rhinitis with cold,heat and normal body constitution were established.Each constitution was divided into intervention group and non intervention group.The cold body intervention group and normal body intervention group were fed with Fuzi Lizhong Decoction,while the heat body intervention group was fed with Huanglian Jiedu Decoction,and the non intervention groups were not given any intervention.On the 0,1,3 and 7 days after the establishment of the model,the content of BMSCs was detected.Results On day 0,the clinical symptom scores of the two cold body constitution groups were significantly higher than those of the two warm body constitution groups and the normal body constitution groups(P all<0.05).There was no significant difference between the normal body constitution group and the warm body constitution group(P all>0.05).Compared with the first day of experiment,the differences in clinical symptom scores on the 1st,3rd,and 7th days of the experiment were not significant in the cold,heat,and normal body non intervention groups(all P>0.05),but the scores on the 7th day of the experiment in the cold,heat,and normal body intervention groups were significantly lower than those on the 1st day of the experiment(all P<0.05).The expression levels of BMSCs on the 1st day,the 3rd day,and the 7th day in the cold body intervention group and normal body intervention group were significantly higher than those in the same constitution non-intervention group at the same time point(all P<0.05);there was no significant difference in the expression of BMSCs between the heat body intervention group and the non-intervention group on the 1st day of the experiment(P>0.05),but the expression in the warm body intervention group was significantly higher than the warm body non-intervention group on the 3rd and 7th day of the experiment(P<0.05).According to the analysis of mixed effects model,the expression level of BMSCs in the non-intervention groups was significantly lower than that in the intervention groups(P<0.05);comparison between different constitutions,the difference in the expression level of BMSCs was not statistically significant between the warm body group and the normal body group(P>0.05);while the expression of BMSCs in the cold body group was significantly less than that of the normal body group and the warm body group(P<0.05).The proliferation trend of BMSCs in the cold,heat,and normal body non intervention group showed a gradual decreasing trend,while the proliferation trend of BMScs in the intervention groups showed a trend of first decreasing and then increasing.Conclusion The proliferation of BMSCs can reflect the constitution of allergic rhinitis.The cold body which is most prone to allergic rhinitis with the least proliferation of BMSCs,and the constitution treatment can promote the proliferation of BMSCs.
