葫芦素B对人MCF-7细胞生物学行为的影响 被引量:1

摘要 目的探讨葫芦素B对激素受体阳性的人MCF-7乳腺癌细胞生物学行为的影响及其机制。方法培养人MCF-7细胞,随机分为对照组和20μM、50μM、100μM葫芦素B组。SRB法观测葫芦素B对人MCF-7细胞增殖活性的影响,Hoechst 33258凋亡实验观察葫芦素B对人MCF-7细胞凋亡的影响,Western Blotting检测自噬相关蛋白。结果与对照组相比,葫芦素B各浓度组能显著抑制人MCF-7细胞增殖,增强其细胞凋亡,差异具有统计学意义(均P<0.05);葫芦素B各浓度组LC3-Ⅱ表达水平显著升高,LC3-Ⅰ、P62表达水平显著下降(均P<0.05)。结论葫芦素B可诱导人MCF-7细胞自噬和凋亡,抑制细胞增殖。 Objective To investigate the effect of cucurbitacin B on the biological behavior of hormone receptor positive human MCF-7 breast cancer cells and its mechanism.Methods Human MCF-7 cells were cultured and randomly divided into control group and cucurbitacin B groups(20μM,50μM,100μM).The effect of cucurbitacin B on the proliferation of MCF-7 cells was observed by SRB method,the effect of cucurbitacin B on the apoptosis of MCF-7 cells was observed by Hoechst 33258 apoptosis assay,and autophagy related proteins were detected by Western blotting.Results Compared with the control group,cucurbitacin B could significantly inhibit the proliferation rate of human MCF-7 cells and enhance their apoptosis(all P<0.05) the expression levels of LC3-Ⅱ in cucurbitacin B groups were significantly increased,while the expression levels of LC3-Ⅰ and P62 were significantly decreased(all P<0.05).Conclusion Cucurbitacin B can inhibit the proliferation of MCF-7 cells and induce autophagy and apoptosis.
