摘要
首先选用60日龄80只生长性能相近的SVF小鼠,随机分为2组,通过腹腔注射方式,对照组注射生理盐水,试验组注射胰岛素,试验期为0.5,1.0,2.0 h以及14 d;14 d后试验则是每48 h注射1次,测定肝脏中有关的microRNA的表达量。接着选用60日龄生长性能相近的10只miR-130a敲除鼠和10只野生型小鼠,随机分为2组,进行胰岛素耐受试验和葡萄糖耐受试验。结果显示,与对照组相比,在注射胰岛素0.5,1.0 h以及连续注射14 d时,miR-146a-5p、miR-143a-3p、miR-125b-2表达显著下调(P<0.05),而miR-130a-3p表达显著上调(P<0.05),在2 h时检测的microRNA均不显著,且在1.0 h时下调与上调的趋势更显著;同时在1.0 h时检测糖类代谢以及脂类代谢相关基因的表达,发现GSK、HK、CD36、FATP1基因表达显著下调,GS基因表达显著上调(P<0.05),但是到了14 d,CD36基因的表达则不存在差异;在敲除miR-130a-3p小鼠中出现胰岛素抵抗与胰岛素耐受的情况。结果表明,miR-146a-5p、miR-143a-3p、miR-125b-2、miR-130a-3p均参与了胰岛素调节的过程,且miR-130a敲除小鼠的胰岛素敏感性降低,为进一步了解胰岛素调节的作用提供有效的试验数据。
First,80 SVF mice with similar growth performance at 60-day-old were randomly divided into two groups.The mice in the control group were injected with saline by intraperitoneal injection,the mice in experimental group was injected with insulin to determine the amount of microRNA expression in the liver,the experimental period was 0.5,1.0,2.0 h and 14.0 d(the injection was performed every 48 h),respectively.Then,10 miR-130 a knockout mice and 1060-day-old wild-type mice with similar growth performance were randomly divided into two groups for insulin resistance test and glucose tolerance test.The results showed that the expression of miR-146 a-5 p,mir-143 a-3 p and miR-125 b-2 were significantly down-regulated(0.5,1.0 h and 14 d after continuous injection)compared with that of the control group(P<0.05),while miR-130 a-3 p expression was significantly up-regulated(P<0.05).There was no significant change in microRNA detected at 2.0 h,but the trend of down-regulation and up-regulation of microRNA was more significant at 1.0 h.The expression of genes related to carbohydrate metabolism in carbohydrate metabolism showed that the expression of GSK,HK,CD36,and FATP1 were significantly down-regulated,and the expression of GS was significantly up-regulated(P<0.05).However,there was no differencein the expression of the CD36 at 14 days.Insulin resistance and insulin tolerance occurred in miR-130 a-3 p knockout mice.It is concluded that miR-146 a-5 p,miR-143 a-3 p,miR-125 b-2,and miR-130 a-3 p are involved in the process of insulin regulation.This study provides effective experimental data for further understanding the role of insulin regulation.
