siRNA干扰ARD1对人结直肠腺癌细胞株基因表达谱的影响

Effect of siRNA interference ARD1 on gene expression profile of human colorectal adenocarcinoma cell line

目的探讨siRNA干扰抑制ARD1的表达对人结直肠腺癌细胞株基因表达谱的影响。方法利用脂质体将筛选出的有效沉默ARD1的siRNA分别转染到SW620、HCT-8两株细胞中,提取两株细胞的总RNA并应用安捷伦人全基因表达谱芯片检测转染前后基因表达谱的变化。结果两株细胞中,转染组相对于阴性对照组,21个基因共同上调2倍以上,15个基因共同下调2倍以上。结论人结直肠腺癌细胞株ARD1沉默前后具有明显不同的基因表达谱,ARD1在结直肠癌发生、发展中发挥原癌基因的作用。

Objective To investigate the effect of siRNA interference on the gene expression profile of human colorectal adenocarcinoma cell line by inhibiting the expression of ARD1.Methods The liposomes were used to transfect the selected siRNA that effectively silenced ARD1 into SW620 and HCT-8 cells.The total RNA of the two cells was extracted and the gene was detected by Agilent human gene expression profile chip Changes in expression profiles.Results Compared with the negative control group,21 genes were up-regulated more than 2 times and 15 genes were down-regulated more than 2 times.Conclusion Human colorectal adenocarcinoma cell line ARD1 is obviously different gene expression profiles before and after silencing,and ARD1 plays a role of proto-oncogene to promote the initiation and development of colorectal cancer.
