富集有核细胞法制备肺癌胸腔积液细胞块中EGFR基因突变的检测

摘要 目的探讨富集有核细胞层沉淀法制备的非小细胞肺癌(non small cell lung cancer,NSCLS)胸腔积液细胞沉渣块中的表皮生长因子受体(EGFR)基因的突变率。方法选取NSCLS伴恶性胸腔积液患者116例,应用富集有核细胞层沉淀法制备胸腔积液细胞沉渣块,并应用突变扩增阻滞系统(ARMS)法检测胸腔积液细胞沉渣块中的EGFR基因的突变情况。结果EGFR基因突变率为50.86%,其中腺癌突变率为57.83%,非腺癌突变率为33.33%。结论富集有核细胞层沉淀法制备的NSCLS胸腔积液细胞沉渣块中EGFR基因突变率高于传统的离心细胞沉渣法,有效提高了肿瘤细胞的获得率及DNA提取率,为肺癌患者的诊断、治疗及分子生物学检测提供了有效手段。 Objective To investigate the detection and analysis of EGFR gene mutations in the pleural effusion cell sediments of non-small cell lung cancer prepared by the enriched nucleated cell layer method.Methods A total of 116 patients with non-small cell lung cancer with metastasis of pleural effusion were collected.The pleural effusion cell sediments were prepared by the enriched nucleated cell layer method and applied ARMS method detects EGFR gene mutations in pleural effusion cell sediments.Results The EGFR gene mutation rate was 50.86%,among which the mutation rate of lung adenocarcinoma was 57.83%,and the mutation rate of non-adenocarcinoma was 33.33%.Conclusion The detection rate of EGFR gene mutation in the pleural effusion cell sediment block of non-small cell lung cancer prepared by the enriched nucleus cell layer method is higher than that of the traditional centrifugal cell sediment method,which effectively improves the rate of tumor cell acquisition and DNA extraction.It will provide effective means for the diagnosis,treatment,and molecular biological detection of lung cancer patients.
