摘要
目的:研究高盐饮食对Dahl盐敏感大鼠肾脏Mcoln3表达的影响。方法:将12只Dahl盐敏感大鼠和12只SS-13BN大鼠分别随机分为正常盐组、高盐组干预3周,采用免疫组化检测肾脏Mcoln3表达位置和表达差异,使用Real-time PCR和Western Blotting方法检测肾脏Mcoln3基因的mRNA和蛋白表达差异。结果:免疫组化显示Mcoln3蛋白在肾脏远端肾小管丰富表达,在高盐负荷后Dahl盐敏感大鼠肾小管Mcoln3蛋白表达明显较正常盐组增加,而13BN-SS大鼠组Mcoln3蛋白变化不明显;同样发现Dahl盐敏感大鼠肾脏Mcoln3的mRNA和蛋白表达在高盐负荷下较13BN-SS大鼠组显著增加,差异具有统计学意义(P<0.05)。结论:我们首次发现肾脏Mcoln3在Dahl盐敏感大鼠在盐负荷后表达异常,可能参与盐敏感性高血压的形成。
Objective:To study the effect of high-salt diet on the expression of Mcoln3 in the kidney of Dahl salt-sensitive rats.Methods:Twelve Dahl salt-sensitive rats and SS-13 BN rats were randomly divided into normal salt group and high-salt group for 3 weeks of intervention.Immunohistochemistry was used to detect the expression position and difference of renal Mcoln3,using Real-time PCR and Western Blotting method were used to detect differences in the mRNA and protein expression of the kidney Mcoln3 gene.Results:Immunohistochemistry showed that Mcln3 protein was abundantly expressed in the distal renal tubules of the kidney.After high salt loading,the Mcoln3 protein expression in the renal tubules of Dahl salt-sensitive rats was significantly increased compared with the normal salt group,while the Mcoln3 protein in the 13 BN-SS rat group did not change;it was also found that the mRNA and protein expression of Mcoln3 in the kidney of Dahl salt-sensitive rats increased significantly compared with the 13 BN-SS rat group under high salt load(P<0.05).Conclusions:We found for the first time that renal Mcoln3 is abnormally expressed in Dahl salt-sensitive rats after salt loading,which may be involved in the formation of salt-sensitive hypertension.
