植物生长调节剂对‘绿岭’核桃组培苗瓶内生根及内源激素含量的影响 被引量:7

Effects of plant growth regulators on endogenous hormone contents and in vitro rooting of‘Lvling’walnut tissue culture seedlings

摘要 为探明核桃组培苗瓶内生根影响因素,以‘绿岭’核桃组织培养继代苗为材料,研究了不同浓度的3-吲哚丁酸钾盐(K-IBA)和吲哚丁酸(IBA)对组培苗瓶内生根及内源激素含量变化的影响。结果表明:外源K-IBA和IBA均对‘绿岭’核桃瓶内生根有促进作用,组培苗培养30~40 d开始形成不定根,K-IBA处理开始生根时间较IBA处理早,根系生长快,但根系细嫩,培养50 d时15 mg/L K-IBA的处理效果最好,其生根率为51.85%、平均根长26.87 mm,均极显著高于其他处理。培养30 d时15 mg/L K-IBA处理的组培苗内源激素IAA、ZT含量最高,分别为221.34、127.50μg/g·FW,均极显著高于其他处理。培养30 d和40 d时15 mg/L K-IBA处理的组培苗内源激素IAA/ZT比值为1.74和2.85、IAA/ABA比值为19.59和14.51,均最高。因此,在生根过程中15 mg/L K-IBA处理可促使生根率提高以及根系的伸长。 To determine the influence factors on in vitro rooting of walnut tissue culture seedlings,tissue culture seedlings of‘Lvling’walnut were used to study effects of different concentrations of 3-indolebutyrate potassium salt(K-IBA)and indolebutyrate on in vitro rooting and contents of endogenous hormone.The results showed that K-IBA and IBA promoted in vitro rooting of'Lvling'walnut tissue culture seedlings,and the adventitious roots formed after being cultured for 30-40 d.Compared with IBA treatment,the tissue culture seedlings of K-IBA treatment rooted earlier,with fast growth rate but more delicate roots.After culturing for 50 d,the treatment of 15 mg/L K-IBA had the best rooting effect,with a rooting rate of 51.85%and an average root length of 26.87 mm,which were significantly higher than other treatments.The contents of endogenous hormones IAA and ZT in tissue culture seedlings treated with 15 mg/L K-IBA for 30 d were the highest,which were 221.34μg/g·FW and 127.50μg/g·FW respectively.After culturing for 30 d and 40 d,the ratios of endogenous hormones IAA/ZT in 15 mg/L K-IBA treatment were 1.74 and 2.85 respectively,and IAA/ABA ratios were 19.59 and 14.51 respectively,which were the highest among all the treatments.Therefore,the treatment of 15 mg/L K-IBA can promote the rooting rate and the elongation of roots.
