摘要
目的探究鼻息肉的病理微环境以及对病理微环境中的免疫细胞是否招募和炎症因子水平变化进行明确,并初步探讨IGG4和STAT5b在鼻息肉中高表达的临床意义。方法收集的临床正常鼻甲和鼻息肉组织进行石蜡和OCT包埋,制作石蜡切片和冰冻切片;采用石蜡切片进行常规病理染色;利用冰冻切片对IGG4与STAT5b在正常鼻甲和鼻息肉中的表达进行免疫荧光定位;提取正常鼻甲和鼻息肉组织蛋白,对IGG4与STAT5b水平进行Western blot检测;同时对鼻息肉病理微环境中的中性粒细胞和巨噬细胞进行免疫荧光检测,并采用PCR检测正常鼻甲和鼻息肉中炎症因子TNF-α,IL-1β,IL-6的mRNA水平。结果相对于正常鼻甲组织,鼻息肉病理组织中呈现纤维化状态,且IGG4与STAT5b水平在鼻息肉中明显升高,此外,鼻息肉病理微环境中的中性粒细胞和巨噬细胞呈现招募趋势,且炎症因子TNF-α,IL-1β,IL-6的mRNA水平显著升高。结论鼻息肉的病理特征呈现纤维化状态,其病理微环境呈现炎症化;鼻息肉中高水平的IGG4与STAT5b可能促进鼻息肉癌变。
Objective To explore the pathological microenvironment of nasal polyps and clarify whether the immune cells are recruited and the changes of inflammatory factor levels in the pathological microenvironment,and preliminarily explore the clinical significance of high expression of IGG4 and STAT5 b in nasal polyps.Methods The collected clinical normal turbinate and nasal polyp tissues were embedded in paraffin and OCT to make paraffin sections and frozen sections;paraffin sections were used for routine pathological staining;frozen sections were used to analyze the expression of IGG4 and STAT5 b in normal turbinates and nasal polyps by immunofluorescence localization;Western blot experiment was performed on the protein extracted from normal turbinate and nasal polyp tissues to quantify the expression of IGG4 and STAT5 b in normal turbinate and nasal polyps;at the same time,immunofluorescence localization was used to visualize the neutrophils and macrophages in the pathological microenvironment of nasal polyps,and PCR was used to detect the changes in the levels of inflammatory factors TNF-α,IL-1β,and IL-6 in normal turbinates and nasal polyps.Results Compared with normal turbinate tissues,the pathological tissues of nasal polyps showed fibrosis,and IGG4 and STAT5 b were highly expressed in nasal polyps.In addition,neutrophils and macrophages in the pathological microenvironment of nasal polyps showed a tendency to recruit.The mRNA levels of inflammatory factors TNF-α,IL-1β,and IL-6 were significantly increased.Conclusion The pathological characteristic of nasal polyps is fibrosis,and the pathological microenvironment is inflammatory.IGG4 and STAT5 b are highly expressed in nasal polyps,which may promote the potential carcinogenesis of nasal polyps.
