不同产地红花指纹图谱建立及其抗氧化活性研究 被引量:5

摘要 通过指纹图谱和聚类分析等方法对15种不同产地红花药材进行比较分析,并对其抗氧化活性进行考察。通过高效液相色谱法(HPLC)等度洗脱来测定15种不同产地红花中羟基红花黄色素A的含量;通过高效液相色谱法梯度洗脱建立15个不同产地红花药材的指纹图谱,以羟基红花黄色素A为指标成分;利用相似度评价及聚类分析进行红花药材产地比较分析;比较分析3种不同提取溶剂对新疆塔城红花药材抗氧化成分的提取效果;采用DPPH方法对15种不同产地红花抗氧化活性进行测试。结果表明:15个不同产地红花中云南红花药材中羟基红花黄色素A含量最高(3.25%),15种产地红花药材除新疆塔城,甘肃,陕西相似度较低外,其他产地红花药材相似度在0.9~1.0之间,说明与标准红花药材存在差异。15个不同产地红花药材最终可聚成2大类:甘肃兰州红花药材和陕西宝鸡红花药材为一类,其他产地红花药材聚为一类。通过用50%甲醇、50%乙醇、50%丙酮对15个不同产地红花进行提取,发现使用50%乙醇提取红花的抗氧化成分提取全面,新疆塔城产地红花药材抗氧化性能力最好。综上,15种不同产地红花指纹图谱及指标成分含量测定可用于全面控制红花药材的质量,15种不同产地红花药材存在差异,对与红花的选用需合理的考量,可以优先考虑道地红花药材。 By means of fingerprint and cluster analysis,15 types of Safflower were compared and analyzed,and their antioxidant activity was investigated.The content of hydroxysafflor yellow A in 15 different producing areas was determined by HPLC,the fingerprint of 15 different producing areas was established by HPLC gradient elution with hydroxy safflower yellow A as the index component.The similarity evaluation and cluster analysis were used to compare the origin of safflower,the extraction effect of three different extraction solvents on the antioxidant components of Xinjiang Tacheng safflower was compared,and the antioxidant activity was tested by DPPH method.The results showed that the content of hydroxysafflor yellow A was 3.25% in Yunnan safflower from 15 different habitats.In addition to Tacheng,Gansu and Shaanxi,the similarity of safflower was low,and the similarity between safflower and standard safflower was between 0.9~1.0.There are two kinds of Safflower in 15 different producing areas:Lanzhou Safflower in Gansu and Baoji Safflower in Shaanxi.By using 50% methanol,50% ethanol and 50% acetone to extract the antioxidant components of safflower from 15 different habitats,50% ethanol was found to have the best effect.In summary,the fingerprint and index content determination of safflower in 15 different producing areas can be used to control the quality of safflower in 15 different producing areas,and the selection of safflower should be considered reasonably.Priority can be given to authentic safflower medicine.
