基于数据库分析INHBA基因在胃癌中的表达及临床意义 被引量:2

摘要 目的筛选并分析胃癌发生发展中INHBA基因的表达及其与临床病理特征和预后的关系。方法通过生物信息学方法分析基因表达综合数据库(GEO)胃癌相关基因芯片GSE79973、GSE13911、GSE19826筛选出差异表达显著的关键基因INHBA。运用GEPIA数据和UALCAN数据库分析INHBA基因在正常胃黏膜组织和胃癌组织的表达差异。运用GEPIA和LinkedOmics在线分析软件分析INHBA基因与胃癌临床病理特征和分期的相关性。然后运用Kaplan-Meier Plotter分析INHBA基因与胃癌患者预后的关系。最后通过STRING数据库分析INHBA基因的蛋白相互作用网络以及可能参与调控的信号通路。结果通过分析三组芯片数据发现,INHBA基因为显著差异基因。数据库分析发现,INHBA在胃癌中明显高表达,且其高表达与胃癌的临床分期和T分期呈正相关。生存分析显示,INHBA高表达的胃癌患者生存预后较差。蛋白相互作用网络分析发现,INHBA可能参与调控TGF-β信号通路从而促进胃癌的发生发展。结论通过数据分析发现,INHAB基因在胃癌中高表达,参与胃癌的发生发展,可能是胃癌潜在的癌基因,有望成为胃癌诊治的重要新靶点。 Objective To screen and analyze the expression of INHBA gene and its relationship with clinicopathological characteristics and prognosis in the gastric cancer by bioinformatics analysis.Methods By analyzing the Gene Expression Comprehensive Database(GEO)gastric cancer related gene expression profiles GSE79973,GSE13911 and GSE19826,the key gene INHBA with significant differential expression was identified.Using GEPIA and UALCAN database to analyze INHBA expression in normal gastric mucosa tissue and gastric cancer tissue.Using GEPIA and Linked Omics online analysis software to analyze the correlation between INHBA gene and clinicopathological characteristics and clinical stage of gastric cancer.Then Kaplan-Meier Plotter was used to analyze the relationship of INHBA expression with the prognosis of gastric cancer patients.Finally,the STRING database was used to analyze the protein interaction network of INHBA and the possible signal pathways involved.Results By analyzing the data of the three microarrays,the INHBA was found to be a significantly upregulated gene.Database analysis found that INHBA was highly expressed in gastric cancer,and its high expression was positively correlated with the clinical stage and T stage of gastric cancer.Survival analysis showed that high expression of INHBA predicted poor clinical prognosis.Protein interaction network analysis showed that INHBA was involved in the regulation of TGF-βsignaling pathway,thereby contributing to the development of gastric cancer.Conclusion INHAB may be a potential oncogene and biomarker of gastric cancer,and is expected to become an important new target for the diagnosis and treatment of gastric cancer.
