基于网络药理学探讨益肾健脾核心药对治疗少弱精症的作用机制

Network pharmacology research of kidney-invigorating and spleen-strengthening core drugs in treatment of oligospermia and asthenospermia

目的:在系统生物学理论的指导下,应用网络药理学的研究方法探讨益肾健脾法治疗少弱精症的物质基础及作用机制。方法:通过检索中药系统药理学平台(TCMSP)选取益肾健脾核心药对中的有效元素和作用靶点;通过检索人类基因信息数据库选取少弱精症的疾病靶点;应用R语言软件遴选药物与疾病共同靶点,应用Cytoscape软件构建药物-疾病-靶点互作网络;通过STRING平台构建共同靶点蛋白互作网络(PPI);应用R语言对益肾健脾核心药物与少弱精症的共同靶点进行GO和KEGG富集分析。结果:从益肾健脾中药中共获得97个有效活性成分,得到46个益肾健脾中药治疗少弱精症的可能靶点,结合KEGG富集分析结果可以推测益肾健脾中药改善男性少弱精症的作用机制可能与调控人内源性激素有关。结论:通过网络药理学的研究方法预测了益肾健脾中药治疗男性少弱精症的可能效应靶点和通路。

Objective:To explore the material basis and mechanism of kidney-invigorating and spleen-strengthening method for treating oligospermia based on network pharmacology.Method:This study selected the effective elements and targets of YishenJianpi core drug pairs by searching the Chinese Medicine System Pharmacology Platform(TCMSP),obatained the disease targets of oligozoospermia by searching the human genetic information database,used R language software to select drugs and disease common target,usedCytoscape software to build a drug-disease-target interaction network,anda common target protein interaction network(PPI)through the STRING platform;R language was used to find the common targets of the core drugs of YishenJianpi and oligospermia and conductenrichment analysis of GO and KEGG.Result:A total of 97 effective active ingredients were obtained from YishenJianpi traditional Chinese medicine,and 46 possible targets for YishenJianpi traditional Chinese medicine to treat oligospermia were obtained.Combined with the results of KEGG enrichment analysis,it can be speculated that YishenJianpi traditional Chinese medicine improved male oligospermia.The whole mechanism of action may be related to the regulation of human endogenous hormones.Conclusion:Through the research method of network pharmacology,the possible effect targets and pathways of YishenJianpi traditional Chinese medicine in the treatment of male oligospermia are predicted.
