摘要
RhoA是一种小的GTP结合蛋白,它的下游效应蛋白Rho激酶(ROCK)调节多种细胞功能,包括细胞生长、基因表达和细胞骨架重组。已有研究表明,这一信号通路在糖尿病并发症、高血压、中风、肿瘤、骨关节炎、急性肺损伤等疾病中具有重要的病理生理作用。糖尿病相关疾病的发生发展与RhoA/ROCK途径的激活或表达上调密切相关,ROCK抑制剂作为药物开发的关键靶点已得到学者的广泛关注。本文就RhoA/ROCK途径与糖尿病相关疾病的关系进行综述,为这一系列疾病的防治提供一种新的、卓有成效的策略。
RhoA is a small GTPase protein.Its downstream effector protein,namely Rho kinase(ROCK),regulates a variety of cell functions,including cell growth,gene expression and cytoskeleton recombination.Studies have demonstrated that this pathway plays an important pathophysiological role in complications of diabetes,hypertension,stroke,tumor,osteoarthritis,acute lung injury,and other diseases.The occurrence and development of diabetes-related diseases are closely related to the activation or up-regulation of RhoA/ROCK pathway.As a key target of drug development,ROCK inhibitors have been widely concerned by scholars.This article reviewed the relationship between RhoA/ROCK pathway and diabetes-related diseases,and offered a new and effective strategy for the prevention and treatment of this series of diseases.
