HPLC法测定百咳静颗粒中8个成分的含量 被引量:15

Simultaneous determination of eight components in Baikejing granules by HPLC

摘要 目的:建立HPLC法同时测定百咳静颗粒中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、橙皮苷、黄芩苷、黄芩素、汉黄芩素8个成分的含量。方法:采用Diamonsil C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速为1.0 mL·min^(-1),柱温45℃,检测波长为215 nm(麻黄碱、伪麻黄碱、苦杏仁苷)、280 nm(甘草苷、橙皮苷、黄芩苷、黄芩素、汉黄芩素),进样量10μL。结果:盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、橙皮苷、黄芩苷、黄芩素、汉黄芩素8个成分的含量测定结果分别为0.312~1.018、0.321~1.175、0.315~0.723、0.876~1.735、0.982~2.604、0.824~9.999、1.635~2.888、0.812~1.661 mg·g^(-1);8个成分均可实现较好的分离,在拟定的浓度范围内,各成分均有良好的线性关系(r≥0.999);平均回收率97.4%~99.7%,RSD为0.65%~1.5%。结论:本方法简便准确,重复性好,可用于百咳静颗粒的质量控制与评价。 Objective:To develop an HPLC method for the simultaneous determination of 8 major components(ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,liquiritin,hesperidin,baicalin,baicalein,wogonin)in Baikejing granules.Methods:The column of Diamonsil C18 with the mobile phase of ethanol-0.1%phosphoric acid at a flow rate of 1.0 mL·min^(-1),the detection wavelength was 215 nm for ephedrine,pseudoephedrine and amygdalin,280 nm for liquiritin,hesperidin,baicalin,baicalein,wogonin,respectively.The temperature of column was 45℃,and the injection volume was 10μL.Results:Determination results of ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,liquiritin,hesperidin,baicalin,baicalein,wogonin were 0.312-1.018 mg·g-1,0.321-1.175 mg·g-1,0.315-0.723 mg·g-1,0.876-1.735 mg·g-1,0.982-2.604 mg·g-1,0.824-9.999 mg·g-1,1.635-2.888 mg·g-1,0.812-1.661 mg·g-1,respectively.Eight components showed good linear relationship within their own ranges(r≥0.999),whose average recoveries were 97.4%-99.7%with RSDs 0.65%-1.5%.Conclusion:The present method is accurate and simple,which can be used for the quality control of Baikejing granules.
