榄绿粗叶木生物碱提取工艺及对小鼠肝癌细胞抑制作用

Extraction Process of Alkaloids from Lasianthus Lancilimbus and Its Inhibitory Effect on Mouse Liver Cancer Cells

摘要 目的优化超声波辅助提取榄绿粗叶木生物碱的最佳工艺,观察榄绿粗叶木生物碱对小鼠肝癌细胞的影响。方法用超声波辅助提取法乙醇浸泡提取榄绿粗叶木的生物碱,利用正交实验的方法优化其提取工艺。CCK-8法检测其生物碱抑制小鼠H22肝癌细胞增殖的效果。通过检测超氧化物歧化酶(SOD)活性,研究榄绿粗叶木生物碱的体外抗氧化活性。结果超声波辅助提取其生物碱的最佳工艺为乙醇浓度70%、料液比1∶45、超声时间60 min、超声功率200 W和超声温度60℃。榄绿粗叶木生物碱对小鼠H22肝癌细胞增殖抑制效果与生物碱浓度和作用时间正相关。榄绿粗叶木生物碱总抗氧化能力与生物碱浓度有明显的量效关系。结论超声辅助法是榄绿粗叶木生物碱提取的有效方法之一,榄绿粗叶木生物碱对小鼠H22肝癌细胞增殖有明显抑制作用,以及榄绿粗叶木具有一定的抗氧化活性。 Objective To optimize the ultrasonic-assisted extraction of the best process of extracting alkaloids of Lasianthus lancilimbus and observe the effect of Alkaloids of Russula alba on mouse liver cancer cells.Methods Ultrasonic-assisted extraction method was used to extract the alkaloids of Ethanol,and the extraction process was optimized by the method of orthogonal experiment.CCK-8 method was used to detect the effect of its alkaloids on inhibiting the proliferation of mouse H22 liver cancer cells.By detecting the activity of superoxide dismutase(SOD),the in vitro antioxidant activity of the alkaloids of the Lasianthus lancilimbus was studied.Results The best technology for ultrasonic-assisted extraction of alkaloids is ethanol concentration of 70%,material-to-liquid ratio of 1:45,ultrasonic time of 60 min,ultrasonic power of 200 W and ultrasonic temperature of 60℃.The inhibitory effect of the alkaloids of Lasianthus lancilimbus on the proliferation of mouse H22 hepatocarcinoma cells was positively correlated with the concentration of the alkaloids and the duration of action.There was a significant dose-effect relationship between the total antioxidant capacity of the alkaloids of Lasianthus lancilimbus and the concentration of alkaloids.Conclusion Ultrasound-assisted method is one of the effective methods for extracting the alkaloids of the Lasianthus lancilimbus.Lasianthus lancilimbus has a significant inhibitory effect on the proliferation of mouse H22 liver cancer cells,and the Lasianthus lancilimbus has certain antioxidant activity.
