摘要 为建立蒙古栎体细胞胚胎发生体系,以蒙古栎未成熟合子胚为外植体,通过不同种源、外植体的取材时间、诱导培养基配比、培养条件等筛选得到胚性愈伤组织,进而进行体胚增殖。结果表明:7月上旬是最佳的取材时期(合子胚大小约10 mm),蒙古栎合子胚在培养基MS+2,4-D 1 mg·L^(-1)+6-BA 0.5 mg·L^(-1)+水解酪蛋白0.5 g·L^(-1)中胚性愈伤组织诱导率可达85.7%,光照条件对胚性愈伤组织的诱导率影响不显著。不同种源对胚性愈伤组织的诱导率有显著影响。在培养基MS+NAA 0.5 mg·L^(-1)+6-BA 0.5 mg·L^(-1)+水解酪蛋白0.5 g·L^(-1)中体胚增殖率最大72.3%,且培养4周时胚性愈伤组织状态最佳。 In this study,immature zygotic embryos were used as explants to establish the somatic embryogenesis system of Quercus mongolica.Embryonal calluses were induced through screening of different factors such as provenances,sampling time of explants,ratio of inducting medium and culture conditions,then somatic embryo proliferation was carried out.The results showed that:early July was the best sampling period(zygotic embryo size was about 10 mm).In MS+1 mg/L 2,4-D+0.5 mg/L 6-BA+0.5 g/L hydrolyzed casein medium,the embryogenic callus induction frequency was up to 85.7%.Light conditions had no significant effect on the induction rate of embryonalcallus.Different provenances had significant influence on the induction rate of embryonalcallus.Somatic embryo proliferation was up to 72.3%in MS+0.5 mg/L NAA+0.5 mg/L 6-BA+0.5 g/L hydrolyzed casein medium,and the embryogenic callus was in the best state after 4 weeks of culture.
