鸟分枝杆菌相关致病基因MAV2054编码蛋白的生物信息学分析

Bioinformatic analysis of the protein encoded by the pathogenic gene MAV2054 related to Mycobacterium avium

摘要 目的运用生物学信息方法分析鸟分枝杆菌相关致病基因MAV2054编码蛋白的生物学特征。方法从NCBI中检索鸟分枝杆菌MAV2054蛋白基因信息。使用序列的fasta格式用在线软件预测MAV2054蛋白理化性质、跨膜区、信号肽、二级和三级结构、T细胞和B细胞表位、亚细胞定位,使用MEGA软件构建系统发育树。结果MAV2054蛋白基因全长924个碱基,共307个氨基酸,其中亮氨酸(Leu)含量高达11.4%,丙氨酸(Ala)9.1%、苏氨酸(Thr)9.1%、无吡咯赖氨酸(Pyl)和硒半胱氨酸(Sec)。该蛋白拥有两个跨膜区(由内向外和由外向内),可能无信号肽。MAV2054蛋白含有3个T细胞表位和28个B细胞表位。二级结构中α螺旋占45%,β折叠15%。三级结构构建成功,可信度96.7%。系统进化树结果显示MAV2054蛋白与MAV2941编码蛋白归一簇,相似度43%。结论生物信息学分析MAV2054蛋白性质较稳定,含有多个T细胞和B细胞优势抗原表位,可作为鸟分枝杆菌肽疫苗研发新靶标。 Objectives To bioinformatically analyze the biological characteristics of the MAV2054 coding protein of Mycobacterium avium.Methods Genetic information on the M.avium MAV2054 protein was retrieved from the NCBI.The FASTA format of the sequence was used to predict the protein’s physical and chemical properties,transmembrane regions,signal peptides,its secondary and tertiary structures,T-cell and B-cell epitopes,and subcellular location of the MAV2054 protein using online software.The software MEGA was used to construct a phylogenetic tree.Results The MAV2054 protein gene is 924 bp in length and it contains a total of 307 amino acids,including leucine(Leu)content as high as 11.4%,alanine(Ala)content of 9.1%,threonine(Thr)content of 9.1%,and no pyrrole-lysine(Pyl)or selenocysteine(Sec).The protein has two transmembrane regions(from the inside to the outside and from the outside to the inside)and may not have a signal peptide.The MAV2054 protein contains 3 T-cell epitopes and 28 B-cell epitopes.αhelices account for 45%of the secondary structure andβsheets account for 15%.The tertiary structure was successfully determined with a reliability of 96.7%.The phylogenetic tree indicated that the MAV2054 protein is divided into two clusters with four other proteins.Conclusion Bioinformatic analysis indicated that the MAV2054 protein is relatively stable,it contains multiple T-cell and B-cell dominant epitopes,and it can be used as a new target for the development of M.avium vaccines.
