白芥子穴位贴敷抑制肝癌H22移植性肿瘤生长的实验研究 被引量:7

摘要 目的:研究白芥子穴位贴敷通过丝裂原活化蛋白激酶(MAPK)途径抑制肝癌H22移植性肿瘤生长的作用及机制。方法:皮下接种H22细胞建立肝癌H22移植性肿瘤小鼠模型,随机分为对照组、白芥子组、白芥子+Ad-NC组、白芥子+Ad-ERK1/2组。白芥子组小鼠给予白芥子穴位贴敷干预;白芥子+Ad-NC组小鼠给予瘤体多点阴性对照腺病毒注射后进行白芥子穴位贴敷干预;白芥子+Ad-ERK1/2组小鼠给予瘤体多点ERK1/2腺病毒注射后进行白芥子穴位贴敷干预,对照组小鼠不进行特殊处理。比较4组小鼠移植瘤重量及磷酸化细胞外调节蛋白激酶1/2(p-ERK1/2)、磷酸化p38(p-p38)、磷酸化c-Jun氨基末端激酶(p-JNK)、细胞周期蛋白D1(cyclinD1)、c-myc、波形蛋白、E-钙黏蛋白表达水平的差异。结果:与对照组比较,白芥子组小鼠移植瘤重量明显减轻,移植瘤中p-ERK1/2、cyclinD1、c-myc、波形蛋白的表达水平明显降低,E-钙黏蛋白的表达水平明显增加(P<0.05),p-p38、p-JNK的表达无明显变化(P>0.05);与白芥子组比较,白芥子+Ad-ERK1/2组小鼠移植瘤重量明显增加,移植瘤中p-ERK1/2、cyclinD1、c-myc、波形蛋白的表达水平明显增高,E-钙黏蛋白的表达水平明显降低(P<0.05),p-p38、p-JNK的表达无明显变化(P>0.05)。结论:白芥子穴位贴敷能够抑制肝癌H22移植性肿瘤生长,抑制MAPK途径中ERK1/2的激活是可能的分子机制。 Objective:To study the inhibitory effect and mechanism of semen brassicae acupoint application on the growth of H22 transplanted tumor via mitogen activated protein kinase(MAPK)pathway.Methods:H22 transplanted tumor model was established by subcutaneous inoculation of H22 cells were,then the mice were randomly divided into control group,semen brassicae group,semen brassicae+Ad NC group,semen brassicae+Ad ERK1/2 group.Semen brassicae group was given semen brassicae acupoint application for intervention,semen brassicae+Ad NC group was given semen brassicae acupoint application for intervention after multiple injections of negative control adenovirus in the tumor,semen brassicae+Ad ERK1/2 group was given semen brassicae acupoint application for intervention after multiple injections of ERK1/2 adenovirus in tumor.The weight of transplanted tumor and the expression of p-ERK1/2,p-p38,p-JNK,cyclin D1,c-myc,vimentin,and E cadherin were compared among the four groups.Results:Compared with the control group,the weight of transplanted tumor significantly reduced,the expression levels of p-ERK1/2,CyclinD1,c-myc,and Vimentin significantly decreased,the expression level of E cadherin significantly increased(P<0.05),while the expression levels of p-p38 and p-JNK did not significantly change(P>0.05)in semen brassicae group;compared with semen brassicae group,the weight of transplanted tumor significantly increased,the expression levels of p-ERK1/2,CyclinD1,c-myc,and Vimentin significantly increased,the expression level of E cadherin significantly decreased(P<0.05),while the expression levels of p-p38 and p-JNK did not significantly change(P>0.05)in semen brassicae+Ad ERK1/2 group.Conclusion:Semen brassicae acupoint application can inhibit the growth of H22 transplanted tumor,and inhibiting the activation of ERK1/2 in the MAPK pathway is a possible molecular mechanism.
