摘要
目的探讨结直肠腺癌中转酮醇酶(transketolase, TKT)的表达及与临床病理特征的关系。方法通过Oncomine和GEO数据库分析TKT基因在结直肠腺癌和癌旁组织中的表达。收集配对结直肠腺癌及癌旁正常肠黏膜组织,分别采用qRT-PCR、Western blot法检测TKT mRNA和蛋白表达,运用免疫组化法检测TKT蛋白表达,并分析其与临床病理特征的相关性。结果 qRT-PCR结果显示:与癌旁组织相比,27例结直肠腺癌组织中TKT mRNA表达显著升高(P<0.001);Western blot结果显示:8例结直肠腺癌组织中TKT蛋白表达高于癌旁组织,在常见结直肠癌细胞株中TKT蛋白表达高于FHC细胞株;两种检测与数据库样本分析一致。免疫组化检测结果显示:92例结直肠腺癌组织中TKT高表达率显著高于癌旁组织(P<0.001),TKT高表达与肿瘤直径(P=0.003)、脉管侵犯(P=0.021)、淋巴结转移(P=0.005)及T分期(P=0.007)相关。结论 TKT在结直肠腺癌中呈高表达,与肿瘤侵袭转移临床病理特征密切相关,其可能参与结直肠腺癌的发生、发展,TKT有望成为结直肠癌预后标志物或潜在的治疗靶点。
Purpose To investigate the expression of transketolase(TKT)and to analyze its correlation with clinicopathologic parameters in colorectal adenocarcinoma.Methods The expression of TKT gene in colorectal adenocarcinoma and para-carcinoma tissues was analyzed using Oncomine and GEO databases.Paired colorectal adenocarcinoma and paracancerous tissues were collected.The transcriptional mRNA level and protein expression of TKT were detected by qRT-PCR and Western blot,respectively.And immunohistochemical staining was used to detect TKT,the results of TKT and its relationship with clinicopathological features were analyzed.Results Compared with the paracancer tissues,the results of qRT-PCR(P<0.001)and Western blot showed that TKT was significantly overexpressed in adenocarcinoma tissues.And the expression of TKT protein in common colorectal cancer cell lines was significantly higher than that in FHC cell lines.The results were consistent with the sample analysis of the above databases.Immunohistochemical results showed that the overexpression rate of TKT in colorectal adenocarcinoma tissues was significantly higher than that in adjacent tissues(P<0.001),and the overexpression of TKT was strongly related to tumor diameter(P=0.003),vascular invasion(P=0.021),lymph node metastasis(P=0.005)and T stage(P=0.007).Conclusion The overexpression of TKT in colorectal adenocarcinoma has a significant correlation with the clinicopathological parameters related to tumor invasion and metastasis.TKT may be a potential prognostic marker or new therapeutic target for colorectal cancer in the future.
