赶黄草活性部位对OGD/R致星形胶质细胞损伤的保护作用及其机制研究

摘要 为探究赶黄草活性部位EPC对OGD/R致星形胶质细胞损伤的保护作用及其初步机制,利用MTT法检测EPC对正常培养和OGD/R后星形胶质细胞活力的影响;Western blot法检测OGD/R损伤的星形胶质细胞内Nrf2、HO-1、ERK1/2、p-ERK1/2、p38、p-p38、JNK1/2和p-JNK1/2蛋白表达水平。结果表明:与空白对照组相比,不同浓度EPC对正常培养的星形胶质细胞均无细胞毒性,且80μg·mL-1 EPC可显著提高星形胶质细胞的活力;同时,与空白对照组相比,OGD/R组细胞中Nrf2、HO-1、p-ERK1/2、p-p38和p-JNK1/2蛋白表达水平显著升高;而与OGD/R组相比,10和20μg·mL-1 EPC可进一步上调Nrf2与HO-1蛋白表达水平,下调p-ERK1/2、p-p38和p-JNK1/2蛋白表达水平,对ERK1/2、p38和JNK1/2蛋白表达水平无影响。这提示EPC对OGD/R致星形胶质细胞损伤具有保护作用,其可能是通过抑制MAPK信号通路进而激活Nrf2/HO-1信号通路来实现的。 The present study aimed to investigate the protective effect of EPC,the active extract of Penthorum chinense,on OGD/R-induced astrocytes and its mechanism.The effects of EPC on the viability of normal and OGD/R-induced astrocytes were detected by a MTT assay.The protein expression levels of Nrf2,HO-1,ERK1/2,p-ERK1/2,JNK1/2,p-JNK1/2,p38and p-p38in OGD/R-induced astrocytes were measured by Western blot.Compared with the blank control group,different concentrates of EPC did not show cytotoxic effects on normal astrocytes,and 80μg·mL-1 of EPC could enhance the cell viability of astrocytes.Moreover,compared with the blank control group,the protein expression levels of Nrf2,HO-1,p-ERK1/2,p-JNK1/2and p-p38in OGD/R-induced astrocytes were significantly increased.Compared with the OGD/R-induced group,10and 20μg·mL-1 of EPC could further upregulate the protein expression levels of Nrf2and HO-1,and downregulate those of p-ERK1/2,p-JNK1/2and p-p38.At the same time,EPC had no effect on those of ERK1/2,JNK1/2and p38.These findings suggest that EPC exhibits the protective activities on OGD/R-induced astrocytes,which may be achieved by blocking the MAPK pathway and further activating the Nrf2/HO-1pathway.
