基于HS-GC-MS的麻桂小儿退热散活性成分的研究

摘要 目的在麻桂小儿退热散临床疗效基础上,采用HS-GC-MS分析鉴定其活性成分。方法采用顶空静态加热萃取技术提取麻桂小儿退热散低温挥发性成分,顶空-气相色谱-质谱(HS-GC-MS)联用技术分析鉴定,Agilent DB-5MS毛细管色谱柱(0.25 m×0.25mm×30m),程序升温,初始温度40℃,保持3min,以4℃/min升至120℃,保持5min,继以8℃/min升至160℃,保持5min;载气为高纯氦气(纯度≥99.999%),载气流量1.0mL/min;分流比100∶1,进样口温度200℃,进样量1μL;质谱鉴定采用电离方式EI,轰击电压70eV,正离子模式,离子源温度230℃。结果从麻桂小儿退热散10批样品得到的HS-GC-MS共有图谱中分析鉴定了20种化合物,其含量之和占挥发油总量的75.33%。结论本研究所分析鉴定挥发性成分作用与临床及文献报道的一致。为临床应用提供了一定的科学依据。 Objective To analyze and identify the active components of Magui Pediatric Antifebrile Powder by headspace sampling-gas chromatography-mass spectrometry(HS-GC-MS). Methods The volatile components of Magui Pediatric Antifebrile powder were extracted by headspace static heating extraction and identified by HS-GC-MS,Agilent DB-5 MS capillary column(0.25 m×0.25 mm×30 m)was used and temperature programmed run was conducted(initial temperature 40℃,hold 3 min,4℃/min rise to 120℃,hold 5 min,8℃/min rise to 160℃,hold 5 min),carrier gas was high purity helium(≥99.999%),carrier gas flow was 1.0 m L/min;splitting ratio was 100∶1,inlet temperature was 200℃ and injection volume was 1μL;mass spectrometry conditions : EI source,bombardment voltage 70 e V,positive ion mode,ion source temperature was at 230℃.Results 20 substances were identified by common chromatogram of HS-GC-MS from 10 batches of samples,accounting for 75.33% of the total volatile oil. Conclusion The analysis and identification of volatile components is consistent with clinical and literature reports. It provides some scientific basis for clinical application.
