代谢组学预测人卵母细胞质量与胚胎体外发育潜能的研究现状 被引量:3

摘要 筛选高质量配子和高发育潜能胚胎是辅助生殖技术关注的重点。代谢物检测是一种非侵入性方法,如果可以应用在配子或胚胎质量的评估中,就可以开发出快速且无创的检测方法,客观地评价配子及胚胎质量,从而有效改善生殖结局。本文总结了代谢物预测卵母细胞质量及胚胎发育潜能的已发表研究,提示在预测配子及胚胎发育的过程中,多参数建模是颇具潜能的方法。现代近红外光谱(NIR)分析技术联合质子核磁共振(NMR)是检测卵泡液和胚胎培养液中代谢物的理想方法。卵泡液中葡萄糖、氨基酸(谷氨酸、缬氨酸)、胆固醇(7β-羟基胆固醇)、维生素(E类)等与卵母细胞质量相关。氧耗(活性氧和氧化应激指标)、氨基酸(谷氨酸和甘氨酸)可能作为预测胚胎发育潜能的标志物。卵泡液中脂质及胚胎培养液中的丙氨酸的预测价值应该进行更深入的研究。 Screening high-quality gametes and embryos with high developmental potential is the key step of current assisted reproductive technology.Metabolomics,as a non-invasive detection method,has great potential in simple and rapid assessment of gamete or embryo quality.In order to identify an optimized metabolomic marker for effective predicting gamete and embryo quality.This paper summarized all the published studies that used metabolites to predict oocyte quality and embryo development potential,which suggested that multi parameter modeling was a potential method for predicting gamete and embryo development.Among the detection methods,near-infrared(NIR)spectroscopy combined with nuclear magnetic resonance(NMR)spectroscopy was an ideal one to detect metabolites in follicular fluid and embryo culture medium.As for the metabolomic indicators,glucose,amino acids(glutamic acid,valine),cholesterol(7β-hydroxycholesterol),and vitamin(E)in follicular fluid are correlated with the quality of oocytes.Oxygen consumption,oxidative stress indicators(such as reactive oxygen species level),and amino acids(glutamic acid and glycine)may be used to predict embryo developmental potential.The predictive value of lipid in follicular fluid and alanine in embryo culture medium should be further studied.
