摘要
目的探究植物乳杆菌BW2013对小鼠肠道微生物和短链脂肪酸的影响。方法 40只BALB/c小鼠随机分为对照组和植物乳杆菌低(2×108 CFU/ml)、中(2×109 CFU/ml)、高剂量组(2×1010CFU/ml),连续灌喂4w后,分别采用实时荧光定量PCR和气相色谱-质谱法(gas chromatography-mass spectrometer,GC-MS)观察不同剂量植物乳杆菌BW2013对健康小鼠肠道微生物和短链脂肪酸的影响。结果中剂量组小鼠体质量在第3w显著增加(P<0.05),第4w极显著增加(P<0.01)。中剂量组小鼠的肠道长度极显著大于对照组(P<0.01)。中、高剂量组小鼠肝体比较对照组无显著差异,各组的肾体比和脾体比与对照组均无明显差异。中、高剂量组双歧杆菌显著高于对照组(P<0.05),而中、高剂量组乳杆菌含量极显著高于对照组(P<0.0001)。低、中、高剂量组丁酸含量极显著高于对照组(P<0.01)。结论中、高剂量的植物乳杆菌BW2013增加小鼠肠道乳杆菌和双歧杆菌的数量;低、中、高剂量的植物乳杆菌BW2013使小鼠肠道丁酸含量极显著增加。植物乳杆菌BW2013具有应用于益生菌保健食品的潜力。[营养学报,2020,42 (5):485-490]
Objective To study the effects of Lactobacillus Plantarum BW2013 on gut microbiota and short-chain fatty acids in mice.Methods Forty BALB/c mice were randomly divided into normal control group and low,medium and high dose groups of Lactobacillus plantarum BW2013(with the dose of 2×108 CFU/ml,2×109 CFU/ml,or 2×1010 CFU/ml,respectively).After 4 weeks of dietary intervention,mice were euthanized and gut microbiota were detected by real-time quantitative PCR,and short-chain fatty acids by gas chromatography-mass spectrometer(GC-MS).Results Compared with the control group,the body weight of the mice in the medium-dose group increased significantly at the third week(P<0.05) and the fourth week(P<0.01).The intestinal length of the mice in the medium-dose group was significantly longer than that of the control group(P<0.01).Compared with the control group,there was no significant difference in the liver/body weight ratio in both the medium and high dose groups,and there were no significant changes in the kidney/body weight or spleen/body weight ratio in the low,medium and high dose groups.The abundance of Bifidobacterium(P<0.05) and Lactobacilleae(P<0.0001) were significantly increased in both the medium and high dose groups.The content of butyric acid in the low,medium and high dose groups was significantly higher than that in the control group(P<0.01).Conclusion Medium and high doses of Lactobacillus plantarum BW2013 increase the abundance of Lactobacillus and bifidobacteria in mice;low,medium and high doses of Lactobacillus plantarum BW2013 significantly increase the intestinal butyric acid content in mice.This study provides an evidence for the application of Lactobacillus plantarum BW2013 in probiotic health food.[ACTA NUTRIMENTA SINICA,2020,42(5):485-490]
