玻璃化冷冻不同阶段小鼠胚胎的比较研究 被引量:4

摘要 目的比较不同阶段小鼠胚胎玻璃化冷冻的存活率以及解冻后的发育潜力。方法运用超促排卵、体外受精得到小鼠的2细胞期胚胎、8细胞期胚胎、桑葚胚、囊胚,分别对其进行玻璃化冷冻、解冻,以未冷冻的胚胎作为对照组。比较各组的冷冻存活率、囊胚形成率及囊胚孵出率。结果 2细胞期胚胎的冷冻存活率、囊胚形成率、囊胚孵出率均低于其他各组,差异均有统计学意义(P<0.05);8 细胞期胚胎的囊胚孵出率低于桑葚胚、囊胚及对照组,差异具有统计学意义(P<0.05);桑葚胚、囊胚及对照组间比较,冷冻存活率、囊胚形成率、囊胚孵出率均无明显差异(P>0.05)。结论 8 细胞期胚胎、桑葚胚、囊胚采用玻璃化冷冻保存的效果优于2细胞期胚胎,桑葚胚、囊胚解冻后的发育潜能更好。 Objective:To compare the post-thaw survival rate and developmental ability of vitrified mouse embryos at various developmental stages.Methods:The 2-cells,8-cells,morula and blastocyst stage mouse embryos were obtained through fertilization in vitro.They were vitrified and thawed to assess their survival and also cultured to determine the rate of blastocyst formation and hatching.The non-vitrified embryos were treated as controls.Results:The survival rate,blastocyst formation rate and hatching rate of vitrified 2-cell embryos were significantly lower than that of 8-cell,morula and blastocyst embryos(P<0.05).The hatching rate of 8-cell embryos was significantly lower than that of morula,blastocysts and non-vitrified embryos (P<0.05).There was no significant difference in the survival rate,blastocyst formation rate and hatching rate among the morula,blastocyst embryos and non-vitrified embryos(P>0.05).Conclusions:Compared to 2-cell embryos,the 8-cell,morula and blastocyst stage mouse embryos had better cryopreservation effect.The embryos vitrified at morula and blastocyst stages would ensure a higher degree of post-warming developmental competence.
