一种检测纽莫康定B_(0)和C_(0)质量浓度的HPLC方法研究 被引量:2

摘要 建立了一种高效液相色谱(HPLC)方法同时测定纽莫康定B_(0)和纽莫康定C_(0)的质量浓度,采用Waters XBridge Amide色谱柱,检测波长220 nm,以V(纯水)∶V(乙腈)为流动相,梯度洗脱。检测结果表明:纽莫康定B_(0)和C_(0)的质量浓度在定量限至400 mg/L范围内线性关系良好,定量限在0.1 mg/L左右,检测限在0.03 mg/L左右,纽莫康定B_(0)和C_(0)的平均回收率分别为99.2%和98.9%,重复性试验纽莫康定B_(0)和C_(0)质量浓度的RSD分别为0.39%和0.42%,纽莫康定B_(0)对照品溶液、纽莫康定C_(0)对照品溶液和供试品溶液在室温条件下24 h内稳定。 An HPLC-based method was established for the simultaneous determination of the mass concentration of B_(0)and C_(0)in pneumocandins.The column of Waters XBridge Amide was used,and the detection wavelength was set at 220 nm.Pure water and acetonitrile were used as mobile phase for gradient elution.The linearity range was LOQs to 400 mg/L,and LOQs was about 0.1 mg/L and LODs was about 0.03 mg/L.The average recoveries of B_(0)and C_(0)were 99.2%with the mass concentration of 0.39%(RSD)and 98.9%with the quality score of 0.42%(RSD).The reference solution and the test solution were stable within 24 hours at room temperature.
