廿烷五烯酸单药和联合用药诱导肺癌细胞凋亡作用原理的研究

摘要 目的:研究廿烷五烯酸在单药和联合用药时对肺癌细胞诱导凋亡作用的可能原理。方法:应用酶标仪检测肺癌细胞的活性氧水平。结果:通过检测肺癌细胞内活性氧(reactive oxygen species,ROS)水平可知,相比卡铂组与对照组而言,EPA组和联合组可明显提高细胞内ROS水平。结论:单独使用廿烷五烯酸或用联合卡铂治疗均可使肺癌细胞内活性氧水平明显升高,从而使bcl-2表达减弱,可能是肺癌细胞凋亡现象增加的原理之一。 Objective:To explore the mechanism of induced apoptosis by eicosapentaenoic acid and carboplatin.Methods:Microplate assay was used to determine Reactive oxygen species(ROS)level of human lung cancer cell line A-549.Results:The level of ROS in EPA group and combined group were significantly higher than those in control group and carboplatin group.Conclusions:ROS lever of EPA group and combined group were obviously increased.expression of bcl-2。That maybe increase the apoptosis of lung cancer cells because ROS can reduce expression of bcl-2.
