藤本月季腋芽离体培养技术研究

Study on In Vitro Culture Technology of Axillary Buds of Morden cvs.of Chlimbers and Ramblers

藤本月季花大而香,是名贵的观赏植物,但采用常规的扦插和嫁接繁殖方法进行繁殖成活率不高,因此,本研究采用藤本月季的腋芽作为外植体进行离体培养以扩大繁殖系数。在初代培养和继代培养阶段分别选用4种培养基配方进行诱导、增殖效果比较。结果表明:藤本月季的腋芽经过消毒,在超净台下切割成0.5 cm,保持生长点,接种到MS+6-BA 1.0 mg/L+NAA 0.5 mg/L培养基上进行初代培养效果最好,待分化出不定芽长至3 cm时再次切割,转入MS+KT 1.0 mg/L+NAA 0.2 mg/L+GA 30.5 mg/L培养基中进行培养,增殖效果最好,且玻璃化现象少。

The flowers of Morden cvs.of Chlimbers and Ramblers are large and fragrant,and it is a precious ornamental plant,but the survival rate of reproduction is not high by using conventional cutting and grafting propagation methods.Therefore,this experiment tried to use the axillary buds of Morden cvs.of Chlimbers and Ramblers as explants for in vitro culture to expand the reproduction coefficient.Four medium formulations were selected in the primary culture and secondary culture stages,through the comparison of induction and proliferation effects.The results showed that the axillary buds of Morden cvs.of Chlimbers and Ramblers were sterilized and cut into 0.5 cm size under a clean table to maintain the growth point.Inoculation to MS+6-BA 1.0 mg/L+NAA 0.5 mg/L medium for primary culture had the best effect.When adventitious shoots grow to 3 cm,they were cut again and transferred to MS+KT 1.0 mg/L+NAA 0.2 mg/L+GA 30.5 mg/L culture medium.The proliferation effect is the best,and the phenomenon of vitrification is less.
