摘要
目的探讨牙龈癌组织中VEGF和PTEN的表达与患者血清中炎性指标水平变化的相关性。方法选择行牙龈癌根治术患者60例作为研究对象,根据其分化程度分为高分化组、中分化组和低分化组,另取癌旁组织作为对照组,免疫组化检测牙龈癌组织中VEGF和PTEN的表达,于术前1 d静脉抽取患者血液,人ELISA试剂盒检测患者血清中炎性因子TNF-α和IL-4的水平,分析牙龈癌组织中VEGF和PTEN的表达与血清中炎性指标水平的相关性。结果(1)与对照组、中分化组和高分化组相比,低分化组VEGF阳性表达率显著增加,PTEN阳性表达率显著下降;中分化组和高分化组PTEN阳性表达率均较对照组显著下降(P均<0.05),其余分组以上两个指标相比差异不具有统计学意义(P均>0.05)。(2)与对照组相比,高、中、低分化组牙龈癌组织中VEGF表达的IOD值和TNF-α含量均显著增加,PTEN表达的IOD值和IL-10含量均显著降低(P均<0.05)。随着分化程度的降低,癌组织中VEGF表达的IOD值和TNF-α含量均显著增加,PTEN表达的IOD值和IL-10含量均显著降低(P均<0.05)。(3)相关性分析结果显示:VEGF表达与IL-10含量呈显著负相关,与TNF-α含量呈显著正相关(P均<0.05);PTEN表达与IL-10含量呈显著正相关,与TNF-α含量呈显著负相关(P均<0.05)。结论牙龈癌患者血清中炎性因子含量与癌组织中VEGF和PTEN的表达具有显著相关性,因此患者血清中炎性因子变化可能与牙龈癌发生、进展相关,也可能一定程度反映患者预后。
Objective To investigate the correlation between the expression of VEGF and PTEN in gingival cancer tissue and the changes of inflammatory indexes in patients'serum.Methods 60 cases of patients underwent radical gingival cancer surgery were selected as the research object,and were divided into high differentiation group,medium differentiation group and low differentiation group according to the degree of differentiation,and another tissue adjacent to cancer was used as the control group.Immunohistochemistry was used to detect the expression of VEGF and PTEN in gingival cancer tissue,and the blood of the patient was drawn intravenously 1 day before the operation,and the levels of inflammatory factors TNF-αand IL-4 in the patient's serum were detected by human ELISA kit.The correlation between the expression of VEGF and PTEN in gingival cancer tissue and the level of inflammatory index in serum was analyzed.Results(1)Compared with the control group,the positive expression rate of VEGF in the poorly differentiated group was significantly increased,and the positive expression rate of PTEN was significantly decreased.The positive expression rates of PTEN in the moderately differentiated group and the well-differentiated group were significantly lower than those of the control group(all P<0.05),the difference between the above two indicators in the other groups is not statistically significant(both P>0.05);(2)Compared with the control group,the IOD value of VEGF expression and TNF-αcontent in gingival cancer tissues of the high,medium and low differentiation groups were significantly increased,and the IOD value of PTEN expression and IL-10 content were significantly reduced(P<0.05);As the degree of differentiation decreased,the IOD value and TNF-αcontent of VEGF expression in cancer tissues were significantly increased,and the IOD value and IL-10 content of PTEN expression were significantly reduced(P<0.05).(3)Correlation analysis results showed that VEGF expression was significantly negatively correlated with IL-10 content,and significantly positively correlated with TNF-αcontent(all P<0.05);PTEN expression was significantly positively correlated with IL-10 content and negatively correlated with TNF-αcontent(all P<0.05).Conclusion The content of inflammatory factors in serum of patients with gingival cancer has a significant correlation with the expression of VEGF and PTEN in cancer tissues.Therefore,changes in inflammatory factors in patients'serum may be related to the occurrence and progression of gingival cancer,and may also reflect the patient's prognosis to a certain extent.
