miR-885-5p和RAC1 mRNA在非小细胞肺癌组织中的表达意义及其相关性 被引量:1

摘要 目的检测微小RNA-885-5p(microRNA-885-5p,miR-885-5p)、ras相关C3肉毒菌毒物底物1(the ras-related C3 botulinum toxin substrate 1,RAC1)mRNA在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达水平,并探讨两者在NSCLC组织中表达的临床意义及其相关性。方法收集2012年3月至2014年7月期间我院诊治的NSCLC患者的癌组织和其配对的癌旁组织110例,Target Scan7.2在线软件预测miR-885-5p与RAC1的结合位点;采用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)检测癌组织和癌旁组织中miR-885-5p、RAC1 mRNA的相对表达量,并分析两者在NSCLC癌组织中表达的相关性;分析NSCLC组织中miR-885-5p、RAC1 mRNA的相对表达量与临床病理参数的关系;Kaplan-Meier生存曲线分析miR-885-5p、RAC1 mRNA的相对表达量对NSCLC患者预后的影响;COX多因素分析影响NSCLC患者独立预后的危险因素。结果miR-885-5p与RAC1基因存在潜在的互补结合位点。与癌旁组织相比,miR-885-5p在NSCLC癌组织中的相对表达量降低(P<0.05),RAC1 mRNA在NSCLC癌组织中的相对表达量增高(P<0.05)。miR-885-5p与RAC1 mRNA在NSCLC癌组织中的相对表达量呈负相关(r=-0.540,P<0.001)。NSCLC癌组织中miR-885-5p的相对表达量与淋巴结转移和TNM分期相关(P<0.05);RAC1 mRNA的相对表达量与肿瘤的分化程度、淋巴结转移和TNM分期相关(P<0.05)。miR-885-5p低表达的NSCLC患者和RAC1mRNA高表达的NSCLC患者预后较差(P<0.05)。miR-885-5p低表达、RAC1 mRNA高表达、TNM分期和淋巴结转移是NSCLC患者不良预后的独立危险因素。结论NSCLC组织中miR-885-5p的表达下调,RAC1 mRNA的表达上调,两者表达呈负相关,且均与预后相关。miR-885-5p、RAC1可能是NSCLC新的预后标志物和治疗靶标。 Objective To detect the expression levels of microRNA-885-5p(miR-885-5p)and ras-related C3 botulinum toxin substrate 1(RAC1)mRNA in non-small cell lung cancer(NSCLC)tissues,and to explore the clinical significance and correlation of their expressions in NSCLC tissues.Methods 110 cases of NSCLC tissues and matched adjacent tissues from NSCLC patients who were treated in our hospital from March,2012 to July,2014 were collected for the study.The binding sites of miR-885-5p with RAC1 were predicted by target scan7.2 software.The relative expression levels of miR-885-5p and RAC1 mRNA in NSCLC and adjacent tissues were detected by quantitative real-time PCR(qRT-PCR),and the correlation between the two expression levels in NSCLC tissues was calculated.The relationship between the expression of miR-885-5p,RAC1 and clinicopathological parameters were analyzed.Kaplan Meier survival curve was applied to analyze the influence of miR-885-5p and RAC1 mRNA expression levels on the prognosis of NSCLC patients.Cox multivariate analysis was performed to analyze the independent prognostic factors of NSCLC patients.Results There were potential complementary binding sites between miR-885-5p and RAC1.Compared with adjacent tissues,the relative expression of miR-885-5p in NSCLC tissues was decreased(P<0.05),while the relative expression of RAC1 mRNA in NSCLC tissues was increased(P<0.05).There was a negative correlation between miR-885-5p and RAC1 mRNA expression in NSCLC tissues(r=-0.540,P=0.028).The expression of miR-885-5p in NSCLC tissues was associated with lymph node metastasis and TNM stages(P<0.05).The expression of RAC1 mRNA in NSCLC tissues was associated with the degrees of differentiation,lymph node metastasis and TNM stages(P<0.05).The prognosis of NSCLC patients with low expression of miR-885-5p and high expression of RAC1 mRNA was relatively poorer(P<0.05).Low expression of miR-885-5p,high expression of RAC1 mRNA,TNM stages and lymph node metastasis were all independent risk factors for the poor prognosis of NSCLC patients.Conclusion The expression of miR-885-5p is down-regulated and RAC1 mRNA is up-regulated in NSCLC.There is a negative correlation between these two,and both of them are related to prognosis of NSCLC patients.miR-885-5p and RAC1 may be used as new prognostic markers and therapeutic targets of NSCLC.
