摘要
目的分析福州地区男同性恋患者中HIV-1病毒CRF_07BC亚型的分子特征和全基因结构。方法采用分层随机抽样法取2011年1月-2016年12月95份福州地区经免疫印迹法确诊的同性恋人群中新近感染的血清标本,用RT-PCR法对env基因进行扩增,通过MEGA软件对env基因序列进化分析,结果表明46份env基因阳性,其中CRF_07BC亚型18株。采用Sanger测序法对CRF07_BC亚型的HIV病毒株(BC.FJFZ.2016.MSM6)进行全基因测序分析。结果福州地区CRF07_BC与云南、河南和江西地区的HIV毒株有较高同源性,序列相似度为95.1%~98.3%。全基因测序结果病毒株(BC.FJFZ.2016.MSM6)是由B亚型和C亚型的重组株,与CRF07_BC标准株(97CN54)的重组位点基本一致。结论HIV-1病毒CRF07_BC亚型已在福州男同人群中传播,导致福州地区MSM中HIV-1流行的新型重组株的流行出现。应加强对男同人群中的CRF07_BC亚型HIV-1病毒的监测与控制。
Objective To analyze the phylogenetic tree based on env gene and Near-whole-genome of HIV-1 virus CRF_07BC subtypes among MSM(men who have sex with men)in Fuzhou.Methods 95 serum samples were selected by stratified sampling,which are from the newly infected HIV-1 MSM confirmed by FZCDC(Fuzhou center for disease control and prevention)with Western Blotting from January 2011 to December 2016.The Env gene was amplified by RT-PCR.After sequencing,the phylogenetic tree of the env gene sequence was analyzed by MEGA software,and 46 env genes were detected,of which 18 were CRF_07BC subtypes.Near-whole-gene analysis of the HIV strain of CRF07_BC subtype(BC.FJFZ.2016.MSM6)was performed by Sanger sequencing.Results CRF07_BC in Fuzhou had higher homology with HIV strains in Yunnan,Henan and Jiangxi,and the sequence similarity was 95.1%~98.3%.The whole gene sequencing result virus strain(BC.FJFZ.2016.MSM6)is a recombinant strain of subtype B and C subtype,which is substantially identical to the recombination site of CRF07_BC standard strain(97CN54).Conclusion The HIV-1 virus CRF07_BC subtype has spreading among MSM in Fuzhou,leading to the prevalence of new recombinant strains.The monitoring and control of the CRF07_BC subtype HIV-1 virus among MSM should be strengthened.
