摘要
目的:研究加减益经汤对环磷酰胺(CTX)所致卵巢功能减退(DOR)大鼠性激素与卵巢组织形态学的影响。方法:成年雌性SD大鼠随机选出10只作为空白对照组,其余大鼠采用CTX 75 mg/kg腹腔单次注射造模,造模成功后将大鼠分为模型对照组、补佳乐组和加减益经汤高、中、低剂量组,每组10只。加减益经汤高、中、低组予加减益经汤,生药剂量分别为23.58、11.79、5.895 g/kg;补佳乐组予补佳乐,0.09 mg/kg;模型对照组与空白对照组灌胃10 mL/kg蒸馏水。造模当日开始予灌胃给药,1次/d,连续28 d。模型对照组在造模后2周,其余各组在给药结束后,于动情间期处死。观察大鼠的一般情况、动情周期、卵巢指数、卵巢组织形态变化,检测血清FSH、LH、E2值。结果:与模型对照组比较,加减益经汤与补佳乐可恢复DOR大鼠动情周期,升高卵巢指数(P<0.01),降低血清FSH、LH(P<0.05或P<0.01),升高血清E2(P<0.01),改善卵巢组织形态,增加原始、成熟卵泡数(P<0.05或P<0.01),降低初级、次级、闭锁卵泡数(P<0.01)。结论:加减益经汤可明显改善CTX所致DOR大鼠的性激素与卵巢组织形态学,其中中剂量是最适宜剂量。
Objective:To study the effect of Modified Yijing Decoction on sex hormone and ovaria n histomorphology in rats with cytoxan(CTX)-induced diminished ovarian reserve(DOR).Methods:Ten adult female SD rats were randomly selected as the blank control group.The remaining rats were modeled by a single intraperitoneal injection of CTX 75 mg/kg.After the model was successfully built,the rats were divided into model control group,Bujiale(补佳乐)group,Modified Yijing Decoction high,medium and low-dose groups,with 10 rats in each group.The Modified Yijing Decoction high,medium and low-dose groups were given Modified Yijing Decoction,the crude drug dosages were 23.58,11.79,5.895 g/kg.The Bujiale group was given Estradiol Valerate Tablets,0.09 mg/kg.The model control group and blank control group was intragastrically administered with 10 mL/kg distilled water.Gavage was started on the day of modeling,once a day,for 28 consecutive days.The model control group was executed 2 weeks after the model was established,and the rest of the groups were sacrificed during the estrus interval after the administration.Observe the rat’s general condition,estrus cycle,ovarian index,ovarian tissue morphology changes,and detect serum FSH,LH,E2 values.Results:Compared with the model control group,Modified Yijing Decoction and Bujiale can restore the estrous cycle of DOR rats,increase ovarian index(P<0.01),and reduce serum FSH and LH(P<0.05 or P<0.01),increase serum E2(P<0.01),improve ovarian tissue morphology,increase the number of primary and mature follicles(P<0.05 or P<0.01),and reduce the number of primary,secondary,and atretic follicles(P<0.01).Conclusion:Modified Yijing Decoction can significantly improve the sex hormones and ovarian tissue morphology of DOR rats caused by CTX,and the medium dose is the most appropriate dose.
