HPLC法同时测定复方川芎片中6种化学成分的含量

Simultaneous Determination of Six Chemical Components in Compound Chuanxiong Tablets(复方川考片)by HPLC

目的:建立复方川芎片中阿魏酸、咖啡酸、川芎嗪、绿原酸、金丝桃苷、藁本内酯6种指标性成分含量检测方法,为市售不同厂家的复方川芎片的质量控制提供科学依据。方法:采用HPLC法同时测定复方中阿魏酸、咖啡酸、川芎嗪、绿原酸、金丝桃苷、藁本内酯6种化学成分的含量,色谱柱为Thermo BDS-C18柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相,柱温为30℃,检测波长为320 nm。结果:阿魏酸、咖啡酸、川芎嗪、绿原酸、金丝桃苷、藁本内酯分离度良好;分别在4.743~94.86μg/mL(r=0.9999)、7.095~141.9μg/mL(r=0.9998)、6.235~124.7μg/mL(r=0.9999)、5.100~102.0μg/mL(r=0.9998)、4.489~89.78μg/mL(r=0.9999)、5.525~110.5μg/mL(r=0.9997)的范围内线性关系良好;平均回收率分别为99.82%(RSD=1.10%)、100.79%(RSD=1.20%)、100.07%(RSD=0.43%)、101.22%(RSD=1.23%)、100.99%(RSD=1.31%)、98.98%(RSD=1.51%)。结论:本研究建立了同时定量分析6种化学成分含量的方法,该法操作方便、准确,便于有效、快速地评价复方川芎片的质量。

Objective:To establish a method for the determination of the content of 6 index components of ferulic acid,caffeic acid,ligustrazine,chlorogenic acid,hyperoside and ligustilide in Compound Chuanxiong Tablets,so as to provide scientific basis for the quality control of Compound Chuanxiong Tablets from different manufacturers.Methods:The contents of ferulic acid,caffeic acid,ligustrazine,chlorogenic acid,hyperoside and ligustilide were determined by HPLC.The chromatographic column was Thermo BDS-C18(4.6 mm×250 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid,the column temperature was 30℃,and the detection wavelength was 320 nm.Results:The resolution of ferulic acid,caffeic acid,ligustrazine,chlorogenic acid,hyperoside and ligustilide displayed good linear relationships within the range of 4.743-94.86μg/mL(r=0.9999),7.095-141.9μg/mL(r=0.9998),6.235-124.7μg/mL(r=0.9999),5.100-102.0μg/mL(r=0.9998),4.489-89.78μg/mL(r=0.9999),5.525-110.5μg/mL(r=0.9999),respectively.The average recoveries were 99.82%(RSD=1.10%),100.79%(RSD=1.20%),100.07%(RSD=0.43%),101.22%(RSD=1.23%),100.99%(RSD=1.31%)and 98.98%(RSD=1.51%)respectively.Conclusion:This study established a method for simultaneous quantitative analysis of the content of six chemical components,the method is convenient and accurate to operate,and is convenient for effective and rapid evaluation of the quality of Compound Chuanxiong Tablets.
