普通烟草WOX转录因子家族的全基因组鉴定及分析 被引量:6

摘要 【背景】WOX(WUSCHEL-related homeobox)转录因子家族是植物特有的一类转录因子家族,被报道参与生长发育、细胞间通讯、干细胞分裂分化稳态调控和器官发育等过程。【方法】利用比较基因组学和生物信息学的方法,在烟草基因组中对WOX转录因子成员进行鉴定,并进行进化分析、共线性分析、保守基序分析以及表达模式等分析。【结果】(1)从烟草基因组中鉴定出23个WOX转录因子家族成员编码基因,其中15个NtWOX基因定位到染色体上;(2)系统发育分析表明,普通烟草和拟南芥的WOX转录因子被分成了3个亚支和9个亚家族;(3)共线性分析发现若干普通烟草NtWOX基因存在于普通烟草与拟南芥基因组的共线性区段中;(4)复制事件分析表明串联重复或者全基因组重复事件在普通烟草WOX家族扩张中起重要作用;(5)保守基序分析表明同一亚家族内NtWOX成员的保守基序的组织形式高度相似;(6)表达分析表明普通烟草NtWOX基因表达具有组织特异性;(7)GO注释分析发现普通烟草NtWOX转录因子参与组织器官发育、细胞间通讯过程和胁迫响应等生物学过程。【结论】本研究在普通烟草中鉴定并系统分析了WOX转录因子家族成员,为其功能研究奠定基础。 [Background]The WOX proteins represent a transcription factor family unique to plants,which play an important role in development,cell-to-cell communication,stem cell homeostasis and organ formation in plants.[Methods]In this study,comparative genomics and bioinformatics methods were used to identify the members of WOX transcription factors in the tobacco genome,and evolutionary analysis,syntenic analysis,conservative motif analysis and expression pattern analysis were performed.[Results]Test data show:1)A total of 23 WOX family members were identified in tobacco by HMMER v3.0,and 15 of which were anchored on chromosomes.2)The phylogenetic analysis revealed that the newly identified tobacco WOX family members together with reported Arabidopsis WOX family members were categorized into 3 clades and 9 subfamilies.3)The evolution relationships of the NtWOX family were further studied via syntenic analysis.4)Several NtWOX genes were predicted to arise from tandem or segmental duplication events,which played an important role in the expansion of the NtWOX family.5)The motif organization within same subgroup were found to be highly conserved.6)The expression pattern analysis showed that the NtWOX genes expression patterns varied with tested tissues.7)The GO analysis indicated the newly identified NtWOX family members might be involved in development,cell-to-cell communication and stress responses.[Conclusion]The results of current study provide insights for the further studies on WOX family members in tobacco.
