藏药四味辣根菜汤散对大鼠心肌缺血再灌注损伤炎症因子的影响

Effects of Tibetan Medicine Siwei Lagencaitang Powder on Inflammatory Factors in Rats with Myocardial Ischemia-reperfusion Injury

目的:探讨藏药四味辣根菜汤散对大鼠心肌缺血再灌注损伤炎症因子的影响。方法:将48只Wistar雄性大鼠随机分为假手术组、模型组、阳性丹参[1.75 g/(kg·d)]组及四味辣根菜汤散低[0.7 g/(kg·d)]、中[1.4 g/(kg·d)]、高[2.1 g/(kg·d)]剂量组。灌胃给药10天后用结扎大鼠左冠状动脉前降支法造心肌缺血再灌注模型,缺血30 min,再灌注90 min,同时观察每只大鼠心电图变化情况。再灌注结束后腹主动脉取血,检测血清中白细胞介素1β(interleukin-1β,IL-1β)、白细胞介素6(interleukin-6,IL-6)、肌红蛋白(myoglobin,Mb)、超敏C反应蛋白(high-sensitivity C-reactive protein,hs-CRP)、肿瘤坏死因子α(tumor necrosis factorα,TNF-α)。结果:与模型组相比,四味辣根菜汤散低、中、高剂量组能降低大鼠血清中IL-1β、IL-6、Mb、hs-CRP、TNF-α含量(P<0.01);与阳性丹参组比较,低剂量组大鼠血清IL-1β和IL-6含量升高(P<0.01),低、中、高剂量组Mb含量及中、高剂量组hs-CRP含量差异有统计学意义(P<0.01),高剂量组大鼠血清TNF-α含量降低(P<0.01)。结论:藏药四味辣根菜汤散对大鼠缺血再灌注损伤导致的炎症具有抑制作用。

Objective:To explore the effects of Tibetan medicine Siwei Lagencaitang powder on inflammatory factors in rats with myocardial ischemia-reperfusion injury.Methods:All 48 male rats were randomized into sham operation group,the model group,Danshen group[1.75 g/(kg·d)],low[0.7 g/(kg·d)],moderate[1.4 g/(kg.d)]and high[2.1 g/(kg·d)]doses groups of Siwei Lagencaitang powder.After drenching for ten days,the models with myocardial ischemia-reperfusion injury were established through ligation of left anterior desending coronary artery in rats,ischemia for 30 minutes,and reperfusion for 90 minutes,simultaneously,to observe ECG changes of each rat.To detect the levels of IL-1β,IL-6,Mb,hs-CRP and TNF-αin the serum when the blood was taken from abdominal aorta by the end of reperfusion.Results:Compared with the model group,Siwei Lagencaitang powder group could lower the contents of IL-1β,IL-6,Mb,hs-CRP and TNF-αin the serum(P<0.01);compared with positive Danshen group,the contents of IL-1βand IL-6 reduced in low dose group(P<0.01),the difference was statistically significant in the contents of Mb in low,moderate and high dosages groups,and the contents of hs-CRP in moderate and high dosage groups(P<0.01),the contents of TNF-αreduced in high dosage group(P<0.01).Conclusion:Tibetan medicine Siwei Lagencaitang powder shows the inhibitory effects on the inflammation induced by myocardial ischemia-reperfusion injury.
