摘要
目的研究不同采样拭子材质和洗脱方式对表面微生物收集的影响,为微生物收集提供依据。方法分别选取3种不同材质的拭子和3种不同洗脱方式下收集伤寒沙门菌、大肠埃希菌及金黄色葡萄球菌,计算细菌的回收率。结果一定菌量负荷后,3种材质拭子结合3种洗脱方式的回收率均超过75%。模拟采样发现,相同的洗脱方式,植绒材质的释放率最高,超声洗脱、旋涡洗脱的回收率均高于70%。应用植绒拭子采样以后经过旋涡振荡1 min,可以取得最佳微生物洗脱效果。结论植绒采样后经涡旋振荡1 min具有最佳的洗脱效果,有利于表面微生物的收集。
Objective To study the effect of different swab materials and elution methods on surface microbial collection,to provide basis for microbial collection.Methods Salmonella typhi,Escherichia coli and Staphylococcus aureus were collected with swabs of three different materials and three different elution methods respectively,calculate the bacterial recovery rate.Results After a certain amount of bacterial load,the recoveries of three swabs combined with three elution methods were all over 75%.Simulated sampling found that,with the same elution method,the flocking material has the highest release rate,the recoveries of ultrasonic elution and vortex elution are both higher than 70%.Sampling with flocking swab was done after a vortex oscillation for 1 minute,the best microbial elution effect can be obtained.Conclusion The best elution effect can be obtained by vortexing oscillation for 1 minute after flocking sampling,it was conducive to the collection of surface microorganisms.
