摘要
目的分析鳄嘴花乙醇提取物不同萃取部位的抗肿瘤活性和化学成分,为鳄嘴花抗肿瘤药物开发奠定前期基础。方法采用WST-1法检测鳄嘴花乙醇提取物的石油醚、二氯甲烷、乙酸乙酯、正丁醇、水层萃取部位及药渣水提取物对人肝癌细胞株(Hep G2)、人胃癌细胞株(MGC803)、人宫颈癌细胞株(He La)细胞的增殖抑制作用;通过高效液相色谱和紫外吸收光谱对各部位进行化学成分分析。结果石油醚部位对Hep G2、He La细胞的抑制率分别为12.3%、40.0%,二氯甲烷部位对Hep G2、MGC803、He La细胞的抑制率分别为30.5%、39.5%、35.7%,乙酸乙酯、正丁醇、水层萃取部位及药渣水提取物对3种肿瘤细胞抑制作用较弱;鳄嘴花主要含植物甾醇、萜类、黄酮类、叶绿素类化合物。结论石油醚部位和二氯甲烷部位的抗肿瘤活性最强,抗肿瘤活性物质为植物甾醇和萜类化合物。
Objective To study the anti-tumor activity and chemical composition of different fractions of ethanol extract of Clinacanthus nutans,and to lay a foundation for the development of anti-tumor drugs from Clinacanthus nutans.Methods WST-1 assay was used to measure the proliferation inhibition rate of HepG2,MGC803 and HeLa cells by the petroleum ether,dichloromethane,ethyl acetate,n-butanol and water fractions of ethanol extract of Clinacanthus nutans and also the water extract of the post-ethanol extraction plant residue.The extraction of ethanol extract of Clinacanthus nutans was chemically analyzed by high performance liquid phase spectrum and ultraviolet absorption analysis.Results The experiments showed that the inhibition rates of the petroleum ether fraction on HepG2,HeLa cells were 12.3%and 40.0%and those of the dichloromethane fraction on HepG2,MGC803 and HeLa cells were 30.5%,39.5%and 35.7%,respectively.The ethyl acetate,n-butanol,water fractions and the water extract of the post-ethanol extraction plant residue had poor inhibitory effects on the three tumor cell.It indicated that Clinacanthus nutans mainly contain Phytosterols,Terpenes,Flavonoids and Chlorophylls.Conclusions The anti-tumor activity of the petroleum ether and dichloromethane fractions is the strongest.The anti-tumor active substances are Phytosterols and Terpenoids.
