枣果实糖组分含量动态变化与相关基因表达分析 被引量:1

摘要 以新疆3个主栽枣品种(骏枣、灰枣及冬枣)在不同发育时期的果实为试验材料,采用高效液相色谱法测定了在果实发育过程中糖组分含量的变化,并应用实时荧光定量PCR法对12个糖代谢关键基因的表达量进行了分析。试验结果表明:3个品种在果实发育过程中糖组分含量的变化规律大致相同,主要糖组分均为果糖、葡萄糖和蔗糖,前期以果糖和葡萄糖积累为主,蔗糖在后期占据主导地位;不同品种中各基因表达量的变化趋势存在一定差异。相关性分析结果显示:基因ZjSPS1对枣果实中蔗糖和还原糖的积累都具有一定的调控作用,与还原糖的积累相关性更强;ZjSS3和ZjSUC2的表达促进了枣果实中还原糖的积累;ZjHK3的表达在还原糖的积累中起抑制作用;ZjvINV1、ZjvINV2和ZjSUC3的低表达在一定程度上促进了蔗糖的积累。 The jujube fruits of Junzao,Grey jujube and Dongzao in Xinjiang at different development stages were selected as test materials in this experiment.During the fruit development process,the content of sugar components was determined by high performance liquid chromatography.The real-time fluorescent quantitative PCR method was used to determine the expression of 12 key genes of sugar metabolism.The results showed that in the three varieties,the changes of sugar components during fruit development were roughly the same.The main sugar components were fructose,glucose and sucrose.Fructose and glucose were mainly accumulated in the early stage,while sucrose was dominant in the late stage.There were some differences in the expression change trends of the genes among different varieties.Correlation analysis showed that ZjSPS1 had a certain regulatory effect on accumulation of sucrose and reducing sugar.It was more relevant to accumulation of reducing sugars.The expression of ZjSS3 and ZjSUC2 could promote accumulation of reducing sugars in jujube fruits.The expression of ZjHK3 played a role in inhibiting accumulation of reducing sugars.The low expression of ZjvINV1,ZjvINV2 and ZjSUC3 promoted the accumulation of sucrose content to a certain extent.
