非小细胞肺癌存活素蛋白和VEGF表达与HPV感染的关系 被引量:5

Correlation between the expression of survivin protein and VEGF in non-small cell lung cancer and HPV infection

摘要 目的探究非小细胞肺癌(NSCLC)存活素(survivin)蛋白、血管内皮生长因子(VEGF)表达及其与人乳头状瘤病毒(HPV)感染的关系。方法选取2018年1月-2019年6月唐山市工人医院收治的100例NSCLC患者为研究对象,采集病例患者新鲜癌组织标本和癌旁组织标本,免疫组化分析NSCLC癌组织和癌旁组织中survivin蛋白、VEGF、HPV表达情况,分析NSCLC患者survivin蛋白、VEGF表达与HPV感染及病理特征的关系。结果 NSCLC患者癌组织survivin蛋白、VEGF、HPV表达阳性率分别为89.00%(89/100)、96.00%(96/100)、39.00%(39/100)高于癌旁组织(P<0.001)。39例HPV感染NSCLC患者survivin阳性15例,survivin阴性24例,HPV感染NSCLC患者survivin阳性组分化程度为低分化、淋巴结有转移的占比分别为53.33%(8/15)、46.67%(7/15)高于survivin阴性组(P<0.05)。39例HPV感染NSCLC患者VEGF阳性19例,VEGF阴性20例,HPV感染NSCLC患者VEGF阳性组临床分期为III期、Ⅳ期、分化程度为低分化、淋巴结有转移的占比分别为71.43%(10/14)、100.00%(1/1)、90.91%(10/11)、88.89%(8/9)高于VEGF阴性组(P<0.05)。NSCLC患者survivin蛋白、VEGF表达阳性率与HPV阳性率呈正相关(P<0.001)。结论 NSCLC患者survivin蛋白、VEGF表达与HPV感染密切相关,HPV感染可能是通过上调survivin蛋白、VEGF的表达,而参与NSCLC患者的恶性生物学行为。 OBJECTIVE To explore the expression of survivin protein and vascular endothelial growth factor(VEGF) in non-small cell lung cancer(NSCLC) and their relationship with human papillomavirus(HPV) infection. METHODS From Jan. 2018 to Jun. 2019, totally 100 patients with NSCLC admitted to the department of laboratory medicine at Tangshan Workers Hospital were selected as the study objects. Fresh cancer tissue specimens and para-cancerous tissue specimens were collected, the expression of survivin protein, VEGF and HPV in NSCLC cancer tissues and paracancerous tissues were analyzed by immunohistochemistry, and the association of survivin protein and VEGF expression with HPV infection and pathological features in patients with NSCLC were also analyzed. RESULTS The positive expression rates of survivin protein, VEGF and HPV in cancer tissues of NSCLC patients were 89.00%(89/100), 96%(96/100) and 39%(39/100), significantly higher than those in para-cancerous tissues(P<0.001). Among the 39 HPV infected patients with NSCLC, there were 15 survivin positive cases and 24 survivin negative cases. The proportions of poor differentiation and lymph node metastasis in HPV-infected NSCLC patients of survivin positive group were 53.33%(8/15) and 46.67%(7/15), respectively, significantly higher than those in survivin negative group(P<0.05). Among the 39 NSCLC patients with HPV infection, there were 19 VEGF positive cases and 20 VEGF negative cases. The proportions of clinical stage III, stage IV, poor differentiation and lymph node metastasis in HPV-infected NSCLC patients of VEGF positive group were 71.43%(10/14), 100%(1/1), 90.91%(10/11) and 88.89%(8/9), respectively, significantly higher than those in VEGF negative group(P<0.05). The positive expression rates of survivin protein and VEGF were positively correlated with the positive rate of HPV in NSCLC patients(P<0.001). CONCLUSION The expression of survivin protein and VEGF in NSCLC patients was closely related to HPV infection. HPV infection may be involved in the malignant biological behavior of NSCLC patients by up-regulating the expression of survivin protein and VEGF.
