摘要
目的肿瘤细胞分选是现阶段实现肿瘤早期诊断的有效手段,微流控芯片技术在细胞分选上具有显著优势,为了设计出一种基于人机交互的肿瘤细胞分选检测技术,引入微流控技术,探究该技术在肿瘤细胞分选、检测中的具体作用效果。方法将微流控芯片技术引入肿瘤细胞分选检测中,利用磁分选技术,通过免疫磁珠靶向标记目标肿瘤细胞,随后结合尺寸分选原理与磁分选技术,制备微流控芯片;在微流控技术对不同肿瘤细胞的混合悬浮液进行细胞的分选、检测的过程中,检测人员通过计算机调整微流控芯片环境中的磁场力大小以及液体流速大小;根据细胞分选检测结果,分析微流控技术的作用效果以及影响因素。结果基于微流控芯片技术的肿瘤细胞分选检测技术,其分选肿瘤细胞的分选效果将受到入口流速、永磁铁剩余磁通密度、细胞悬浮液流速等因素的影响。人为控制这些影响因素,进行cell-MNPs和HL-7720等两种细胞的分选,当细胞混悬液的流速为50uL/min时,目标细胞cell-MNPs分选率达68.3%。结论制备的微流控芯片能够完成肿瘤细胞的分选、检测实验,对肿瘤细胞的检测、分选效率较好,分选过程不受细胞惯性力的作用,因此可将其作为肿瘤分选检测系统的主要技术模块。
Objective Tumor cell sorting is an effective means to achieve early diagnosis of tumor at this stage.Microfluidic chip technology has significant advantages in cell sorting.In order to design a kind of tumor cell sorting detection technology based on human-computer interaction,microfluidic technology is introduced to explore the specific effect of this technology in tumor cell sorting and detection.Methods The microfluidic chip technology was introduced into the separation and detection of tumor cells.Using magnetic separation technology,the target tumor cells were labeled by immunomagnetic beads,and then the microfluidic chip was prepared by combining the size separation principle and magnetic separation technology.In the process of sorting and detecting the mixed suspension of different tumor cells by microfluidic technology,the detection personnel adjusted the microfluidic chip by computer According to the results of cell sorting,the effect and influencing factors of microfluidic technology were analyzed.Result Based on microfluidic chip technology,the sorting effect of tumor cells will be affected by inlet flow rate,residual magnetic flux density of permanent magnet,flow rate of cell suspension and other factors.When the flow rate of cell suspension was 50 UL/min,the sorting rate of cell MNPs was 68.3%.Conclusion The microfluidic chip can complete the sorting and detection experiments of tumor cells,and has good detection and sorting efficiency.The sorting process is not affected by the inertial force of cells,so it can be used as the main technical module of tumor sorting and detection system.
