同源异型盒A5、高迁移率族蛋白1在卵巢癌中表达及临床意义

摘要 目的探讨同源异型盒A5(HOXA5)、高迁移率族蛋白1(HMGB1)在卵巢癌中的表达及临床意义。方法分别取38例上皮性卵巢癌患者、22例良性上皮性卵巢肿瘤患者、20例无卵巢病变患者的卵巢癌组织、良性病变组织、正常组织,免疫组化法检测不同组织中HOXA5、HMGB1蛋白的表达情况,及其与患者临床特征的关系;采用逆转录聚合酶链反应(RT-PCR)检测不同组织中HOXA5、HMGB1 mRNA的相对表达量。结果良性病变组织和卵巢癌组织中HOXA5蛋白的阳性表达率和HOXA5 mRNA的相对表达量均低于正常组织,卵巢癌组织中HOXA5蛋白的阳性表达率和HOXA5 mRNA的相对表达量均低于良性病变组织,差异均有统计学意义(P﹤0.05)。卵巢癌组织中HMGB1蛋白的阳性表达率和HMGB1 mRNA的相对表达量均高于良性病变组织和正常组织,差异均有统计学意义(P﹤0.05)。不同淋巴结转移情况、组织学分级卵巢癌患者卵巢癌组织中HOXA5、HMGB1蛋白阳性表达率比较,差异均有统计学意义(P﹤0.05)。Pearson相关性分析显示,卵巢癌组织中,HMGB1表达与HOXA5呈负相关(r=-0.371,P﹤0.05)。结论HOXA5在卵巢癌组织中低表达,HMGB1高表达,二者异常表达可能与卵巢癌组织的分化、淋巴结转移有关。 Objective To explore the expression and clinical significance of home box A5(HOXA5)and high mobility group box 1(HMGB1)in ovarian cancer.Method Take 38 patients with epithelial ovarian cancer,22 patients with benign epithelial ovarian tumors,and 20 patients with ovarian cancer,benign lesions,and normal tissues.Immunohistochemical method to detect the expression of HOXA5 and HMGB1 proteins in different tissues and their relationship with the clinical characteristics of patients.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the relative expression of HOXA5 mRNA and HMGB1 mRNA in different tissues.Result The positive expression rate of HOXA5 protein and the relative expression of HOXA5 mRNA in benign lesion tissues and ovarian cancer tissues were lower than those in normal tissues,the positive expression rate of HOXA5 protein and relative expression of HOXA5 mRNA in ovarian cancer tissues were lower than those in benign lesion tissues,the differences were statistically significant(P<0.05).The positive expression rate of HMGB1 protein and the relative expression of HMGB1 mRNA in ovarian cancer tissues were higher than those of benign diseased tissues and normal tissues,and the differences were statistically significant(P<0.05).The comparison of the positive expression rates of HOXA5 and HMGB1 protein in ovarian cancer tissues of patients with different lymph node metastasis and histological grades was statistically significant(P<0.05).Pearson correlation analysis showed that,in ovarian cancer,the expression of HMGB1 was negatively correlated with HOXA5(r=-0.371,P<0.05).Conclusion HOXA5 is lowly expressed in ovarian cancer tissues while HMGB1 is highly expressed.The abnormal expression of both targets may be related to the differentiation of ovarian cancer tissues and lymph node metastasis.
