摘要
目的观察远志皂苷对转tau基因果蝇阿尔茨海默病模型行为学及寿命的影响。方法将待测果蝇分为3组:空白组(正常果蝇喂基础培养基)、模型组(转tau果蝇喂基础培养基)、远志组(转tau果蝇喂2%浓度远志皂苷培养基)。5 d后对3组果蝇的攀附指数、进入无障碍离心管个数、平均寿命、最高寿命及半数死亡时间进行比较。结果远志皂苷组攀附指数显著高于模型组(P<0.05);远志皂苷组进入无障碍离心管个数显著高于模型组(P<0.05);远志皂苷组半数死亡时间、平均寿命、最高寿命显著高于模型组(P<0.05)。结论远志皂苷能提高转tau基因果蝇阿尔茨海默病模型的运动能力、学习记忆能力,延长其寿命。
Objective To observe the effect of Polygala saponins on the behavior and lifespan of the tau-transgenic Drosophila model of Alzheimer’s disease.Methods The Drosophila melanogasters in the study were divided into three groups:blank group (normal Drosophila melanogasters fed with basic medium),model group (tau-transgenic Drosophila melanogasters fed with basic medium),Polygala group (tau-transgenic Drosophila melanogasters fed with 2% Polygala saponins medium).Five days later,the climbing index,the number of barrier-free centrifuge tubes,the average lifespan,the maximum lifespan and the half-death time of the three groups were compared.Results The climbing index of Polygala saponins group was significantly higher than that of the model group (P<0.05).The number of Drosophila melanogasters entering the barrier-free centrifugal tube in the Polygala saponin group was significantly higher than that in the model group (P<0.05).The half-death time,average lifespan and maximum lifespan in the Polygala saponins group were significantly higher than those in the model group (P<0.05).Conclusion Polygala saponins can improve the motor ability,learning and memory ability and prolong the lifespan of tau-transgenic Drosophila model of Alzheimer’s disease.
