盐酸右美托咪定通过调控miR-223对子宫内膜癌细胞增殖和凋亡的影响

Effect of dexmedetomidine hydrochloride on proliferation and apoptosis of endometrial cancer cells by regulating miR-223

摘要 目的探讨盐酸右美托咪定(Dex)对子宫内膜癌细胞AN3CA增殖和凋亡的影响及作用机制。方法AN3CA细胞分为对照组、不同浓度(1、2、5μmol/L)Dex组、miR-223组、miR-NC组、5μmol/L Dex+anti-miR-223组和5μmol/L Dex+anti-miR-NC组。MTT法检测细胞增殖,流式细胞仪检测细胞凋亡,Western Blot法检测细胞中CyclinD1、P21、Bcl-2和Bax蛋白表达水平,RT-qPCR法检测miR-223表达水平。结果与对照组比较,Dex组AN3CA细胞活性及CyclinD1、Bcl-2蛋白水平降低(P<0.05),细胞凋亡率、P21和Bax蛋白及miR-223水平升高(P<0.05)。与miR-NC组比较,miR-223组AN3CA细胞活性及CyclinD1、Bcl-2蛋白水平降低(P<0.05),细胞凋亡率及P21、Bax蛋白水平升高(P<0.05)。与5μmol/L Dex+anti-NC组比较,5μmol/L Dex+anti-miR-miR-223组AN3CA细胞活性及CyclinD1、Bcl-2蛋白水平升高(P<0.05),细胞凋亡率及P21、Bax蛋白水平降低(P<0.05)。结论Dex可能通过上调miR-223降低子宫内膜癌细胞AN3CA增殖,并诱导细胞凋亡。 Objective To investigate the effects and mechanism of dexmedetomidine hydrochloride(Dex)on proliferation and apoptosis of endometrial cancer cell line AN3CA.Methods AN3CA cells were divided into control group,Dex groups of different concentrations(1,2,5μmol/L correspongdingly),miR-223 group,miR-NC group,5μmol/L Dex+anti-miR-223 group,5μmol/L Dex+anti-miR-NC group.MTT was used to detect cell proliferation,flow cytometry was used to detect apoptosis,Western blot was used to detect expression levels of CyclinD1,P21,Bcl-2 and Bax protein,and RT-qPCR was used to detect expression level of miR-223.Results Compared with the control group,AN3CA cell activity,the protein levels of CyclinD1 and Bcl-2 decreased in Dex groups(P<0.05),while the apoptosis rate,the protein levels of P21 and Bax,and miR-223 levels increased(P<0.05).Compared with the miR-NC group,AN3CA cell activity,the protein levels of CyclinD1 and Bcl-2 decreased in the miR-223 group(P<0.05),while the apoptosis rate and the protein levels of P21 and Bax increased(P<0.05).Compared with the 5μmol/L Dex+anti-NC group,AN3CA cell activity and the protein levels of CyclinD1 and Bcl-2 increased in the 5μmol/L Dex+anti-miR-223 group(P<0.05),while the apoptosis rate and the protein levels of P21 and Bax decreased(P<0.05).Conclusion Dex may inhibit the proliferation of endometrial cancer cell AN3CA and promote its apoptosis by up-regulating miR-223.
