腊梅叶醇提物二氯甲烷部位对阿尔茨海默病小鼠的影响 被引量:4

Effects of The Dichloromethane Fraction in The Phytol Extract of Chimonanthus Praecox on Alzheimer'S Disease Mice

摘要 目的:探讨腊梅叶醇提物二氯甲烷部位对阿尔茨海默病(alzheimer's disease,AD)小鼠的影响与机制。方法:皮下大剂量注射D-半乳糖联合东莨菪碱腹腔注射建立AD小鼠模型,经腊梅叶醇提物二氯甲烷部位高、中、低(350、175、87.5mg·kg^(-1))剂量组和阳性对照药吡拉西坦干预后,测定小鼠找到平台的潜伏期及穿过平台的次数,测定各组小鼠脑重指数,检测脑组织、肝组织、超氧化物歧化酶(Superoxide Dismutase,SOD)、丙二醛(Malondialdehyde,MDA)含量,测定脑组织乙酰胆碱酯酶(acetylcholinesterase,AChE)、乙酰转移酶(acetyltransferase,ChAT)的含量。结果:腊梅叶醇提物二氯甲烷部位能改善AD小鼠的学习、记忆能力,提高小鼠脑组织、肝组织、血清的SOD,降低MDA的活性,提高小鼠脑组织的ChAT活性,降低AChE的活性(P<0.05)。结论:腊梅叶醇提物二氯甲烷部位能够改善AD小鼠的学习记忆能力,其作用机制可能与提高SOD、ChAT,降低MDA、AChE的含量有关。 Objective:It is to explore the impact and mechanism of Chimonanthus Praecox on mice with Alzheimer's Disease(AD).Methods:After the intervention of three dose groups(high dose group:350 mg·kg^(-1);middle dose group:175 mg·kg^(-1),and low dose group:87.5mg·kg^(-1))of the dichloromethane fraction of the phytol extract of Chimonanthus Praecox and the positive control drug Piracetam,the Morris Water Maze was adopted to measure and find the incubation period of the mice platform as well as the times of passing through the original platform;the brain weight index of each group was measured;the contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in brain tissue,liver tissue,and serum were detected by UV-Vis spectrophotometer,and the contents of acetylcholinesterase(AChE)and acetyltransferase(ChAT)in brain tissue were measured by the same method.Results:The fraction of dichloromethane,the phytol extract of Chimonanthus Praecox,can improve the learning,memory,and cognitive ability of AD mice.It can increase SOD and ChAT,and it also can decrease the contents of MDA,AChE.Conclusion:The fraction of dichloromethane,the phytol extract of Chimonanthus Praecox,can improve the learning,memory,and cognitive ability of AD mice.The mechanism of the dichloromethane fraction in the phytol extract of Chimonanthus Praecox might be related to the increase of SOD and ChAT,the decrease of the contents of MDA,AChE.
