矮牵牛PhLcyB基因的克隆及功能分析 被引量:2

Cloning and Functional Analysis of PhLcyB Gene from Petunia hybrida

摘要 本研究以矮牵牛为实验材料,利用VIGS技术验证矮牵牛类胡萝卜素代谢途径关键酶基因PhLcyB的功能,为探究植物叶片中类胡萝卜素的代谢调控的分子机制及类胡萝卜素在叶片呈色方面的作用机理提供理论依据。本实验克隆获得了PhLcyB基因的完整ORF序列,通过VIGS技术构建了TRV-PhLcyB和TRV-PhCHS重组载体,通过侵染发现TRV-PhLcyB侵染的的矮牵牛植株叶片出现大面积白化现象,花瓣颜色没有明显变化。进行叶片生化检测后,发现PhLcyB基因沉默后叶片中类胡萝卜素含量明显降低,尤其是叶黄素降低明显,同时叶片中叶绿素a和叶绿素b含量明显降低。PhLcyB基因对矮牵牛叶片中类胡萝卜素(尤其是叶黄素)的代谢调节有重要作用。 In this study,Petunia hybrida was used as experimental material to verify the function of PhLcyB,a key enzyme gene in the carotenoid metabolism pathway of Petunia hybrida,and to provide a theoretical basis for exploring the metabolic regulation mechanism of carotenoids and the role of carotenoids in color formation.The complete ORF sequence of PhLcyB gene was cloned in this experiment.The recombinant vector of TRV-PhLcyB and TRV-PhCHS was constructed by VIGS technology.The infection of TRV-PhLcyB infected leaves of Petunia hybrida showed large-area whitening.There is no significant change in color.After biochemical detection,it was found that the content of carotenoids in leaves was significantly decreased after the PhLcyB gene was silenced,especially the lutein was significantly decreased,and the contents of chlorophyll a and chlorophyll b in leaves were significantly decreased.The PhLcyB gene plays an important role in the metabolic regulation of carotenoids(Especially lutein)in petunia leaves.
